改良伸直位固定法建立兔膝骨性关节炎模型的研究 被引量:16

摘要 目的用改良伸直位固定法建立兔膝骨性关节炎动物模型,为艾灸防治早期膝,骨性关节炎提供动物模型。方法 24只日本大耳白兔随机分为改良伸直位固定纽(A组)、伸直位固定组(B组)和正常组(C组),每组8只。A组用改良法固定兔膝关节于伸直位;B组管型石膏固定兔膝关节伸直位:C组不做固定正常喂养。在造模8周后,各组分别从肉眼、光镜下、透射电镜下观察兔膝关节组织形态学情况,并用Mankin评分比较各组膝关节软骨损伤情况。结果通过肉眼、光镜、电镜下观察,A、B纽均呈现OA典型病理改变,在Mankin评分上:A、B组明显高于C组,比较均有有非常显著意义(f=21.177,P<0.01;f=32.233,P<0.01);A组高于B组,但比较无显著意义(t=1.342,P>0.05)。结论改良伸直位固定法能成功建立兔膝0A模型,且本造模法具有简单、固定牢固、无创伤和造模成功率高等优点,有利于研究艾灸疗法对兔膝OA的防治作用。 Objective To establish rabbit's model of knee osteoarthritis(OA)with modified method of immobilization in extension position,which could be used in observation of therapeutic and protective effect of Moxibustion on OA of rabbits.Method Twenty four Japanese white rabbits were divided into three groups at random(8 rabbits in each group):group A,group B,group C,the left hind limb of animals in group A were immobilized with modified method in extension position,group B were immobilized with plaster cast in extension position,and group C was used for normal control.After 8 weeks,morphological and histological changes of each group were evaluated through unaided eyes,optical microscope,transmission electron microscopic(TEM)and Mankin' score by examination the appearance of cartilage.Results The foundings of histological study showed that changes in both group A and group B accorded with character of OA,and Mankin' scores in group A and B were higher than in group C respectively(f=21.177,P0.01;f=32.233,P0.01);the scores in group A were not significantly different from in group B(t=1.342,P0.05).Conclusion The method of modified with immobilization in extension position can successfully induce the rabbit's model of knee OA,and has is advantages of simplicity,sturdy fixation,non-invasion and higher succeed rate,which is in favor of observation of therapeutic and protective effect of Moxibustion on OA of rabbits.
