摘要
目的:观察骨刺消巴布剂对大鼠急性软组织损伤细胞凋亡和凋亡调控基因Bcl-2及其抑制基因Bax的影响,探讨骨刺消巴布剂对急性软组织损伤细胞的保护作用是否与抑制细胞凋亡及调节Bcl-2、Bax的表达有关。方法:选用SPF级SD大鼠40只,随机分成4组:骨刺消巴布剂组、奇正消痛贴组、空白对照组、正常组。骨刺消巴布剂组、奇正消痛贴组、空白对照组,通过局部打击来制备大鼠急性软组织损伤模型,骨刺消巴布剂组、奇正消痛贴组外敷相应药物,空白对照组、正常组不做任何处理,给药3周后处死动物,TUNEL法检测软组织损伤细胞的凋亡指数,免疫组化法检测组织细胞中Bcl-2、Bax水平。结果:骨刺消巴布剂组、奇正消痛贴组细胞凋亡率明显低于空白对照组(P<0.05),Bcl-2在骨刺消巴布剂组、奇正消痛贴组中的表达高于空白对照组(P<0.05),Bax的表达低于空白对照组(P<0.05),骨刺消巴布剂组、奇正消痛贴组比较无统计学意义。结论:Bcl-2、Bax是细胞凋亡的2个调控基因,骨刺消巴布剂对损伤的软组织细胞的影响机制可能是通过上调Bcl-2的表达,下调Bax的表达来抑制细胞的过度凋亡,这可能是该药治疗急性软组织损伤的机制之一。
Objective: To observe the effects of GuCiXiao cataplasm on cellular apoptosis of the rats with acute soft tissue injury, apoptosis regulation gene Bcl-2 and the inhibiting gene Bax, in order to discuss whether the protection of GuCiXiao cataplasm to the injured cells was related to the expressions of inhibiting cellular apoptosis and regulation genes Bcl-2 and Bax. Method: All 40 rats were randomized into four groups: GuCiXiao cataplasm group, QiZheng XiaoTong Tie group, blank control group and normal group. Except the normal group, the rat models in others groups were established by local beat, GuCiXiao cataplasm group and QiZheng XiaoTong Tie group were administered with drugs for external use, the other groups were given with nothing, after treating for three weeks, the animals were put to death, apoptosis index of the cells in rats with soft tissue injury was detected with TUNEL method, the level of Bcl-2 and Bax of the cells in the tissue were tested by immunohistochemistry. Result: Cellular apoptosis rates of GuCiXiao cataplasm group and QiZheng XiaoTong Tie group were lower than the rate of blank control group obviously (P〈0.05), the expressions of Bcl-2 in both groups were higher than that of blank control group (P〈0.05), the expressions of Bax were lower than that of blank control group (P〈0.05), the comparisons between GuCiXiao cataplasm group and QiZheng XiaoTong Tie group demonstrated no statistical meaning. Conclusion: One of the mechanisms for GuCiXiao cataplasm in treating acute soft tissue injury might be through upregulating the expressions of Bel and reducing the expressions of Bax to inhibit excessive apoptosis of the cells, Bcl and Bax are the genes regulating cellular apoptosis.
