摘要
目的:通过检测DNA损伤标志物8-氧鸟嘌呤脱氧核苷(8-oxo-dG)在甲型H1N1型流感病毒感染MDCK细胞中的表达,初步探讨甲型H1N1型流感病毒诱导细胞凋亡的脱氧核糖核酸(DNA)损伤机制。方法:MDCK细胞培养后,甲型H1N1流感病毒感染继续培养0h、1h、3h、6h、12h、24h、48h,细胞爬片免疫组织化学方法检测各时间点DNA损伤标志物8-oxo-dG的表达。结果:甲型H1N1流感病毒感染后各时间点实验组MDCK细胞8-oxo-dG的表达均显著高于相应的正常对照组(P<0.05),病毒感染各组间两两比较发现各组间差异显著。结论:甲型H1N1流感病毒感染能造成宿主细胞DNA氧化损伤。
Objective:To explore the apoptosis mechanism of H1N1 influenza virus infection to MDCK cell by detecting the expression of 8-hydroxy-2'-desoxyguanosine(8-oxo-dG) as biochemical marker of DNA damage.Methods:MDCK cells were seeded in slide of 48-well plate and infected with H1N1 influenza virus.Immunohistochemistry method was used to investigate 8-oxo-dG expression in MDCK cell respectively after being infected for 0,1,3,6,12,24 and 48 hours.Results:Significant difference of 8-oxo-dG in MDCK cells could be observed between every case-control pair at 0,1,3,6,12,24 and 48 hours;and significant increase of 8-oxo-dG could also be found among the experiment groups(P0.05).Conclusion:H1N1 influenza virus infection can induce significant DNA oxidative damage.
