摘要
目的观察H1N1型流感病毒诱导的MDCK细胞抗氧化相关修复基因MTH1表达量的变化。方法 MDCK细胞培养后,H1N1型流感病毒感染0、1、3、6、12、24、48h,在各时间点经RT-PCR反应检测细胞内MTH1基因的相对表达量。结果甲型H1N1流感病毒感染后0、6、12h,MTH1基因的表达量与正常对照组差异无统计学意义;感染后1、3hMTH1基因的表达量显著高于正常对照组;感染后24、48h表达量显著低于正常对照组。结论 H1N1流感病毒感染细胞的MTH1基因表达的增加,可能增加细胞对DNA氧化损伤的修复能力,在流感发生和防御机制中起到作用。
Objective To observe the expression changes of antioxidant-retated repair genes MTH1 in the H1N1 influenza virus infected MDCK cell. Methods MDCK cells were cultured and infected with H1N1. The relative expression of MTH1 gene of the host cells were detected by the RT-PCR after being infected for 0,1,3,6,12,24 and 48 hours. Results There were no significant differences of the MTH1 gene expression between the experimental groups and the control groups for the 0, 6 and 12 hours,but was significantly higher than the control group after infected for 1 h,and 3 h,and expression was significantly lower than the control group after infected for 24 h, and 48 h. Conclusion The expression of MTH1 gene H 1N1 was increased in influenza virus infected cells.Increased expression of MTH1 gene may increase the cells ability to repair DNA oxidative damage. This may play a role in defense of influenza.
