Establishment and optimization of SSR-PCR reaction system in flax

摘要 通过研究SSR-PCR反应体系的主要成分对扩增结果的影响,建立了亚麻SSR反应的优化体系,即总体积20μL,Mg2+1.9mmol.L-1,dNTPs 0.55mmol.L-1,Taq DNA聚合酶1.5U,25ng.μL-1引物75ng,DNA模板(50ng.μL-1)100ng.利用10个亚麻材料对优化后的SSR体系进行验证,1.5%琼脂糖检测结果显示,优化后的SSR体系稳定性,重复性好,扩增效果良好. The factors which affected the SSR-PCR reaction results were studied in the experiment,the optimum SSR reaction system of flax was established,it was showed as follows:total volume of reaction was 20 μL,the optimum concentrations of Mg^2+ and dNTPs were 1.9 mmol·L^-1 and 0.55 mmol·L^-1 respectively,moreover,1.5 U Taq DNA polymerase,50 ng·μL^-1 template DNA 100ng,25 ng·μL^-1 primer 75 ng.To evaluate the SSR system,10 different flax germplasm were used and tested by 1.5 % agarose gel,the PCR results showed that this SSR reaction system was steady and reproducible.
