摘要
本文采用荧光探针的方法,通过测量与紫膜结合后的荧光探针ANS的荧光寿命随介质pH的变化,研究了介质pH对bR的构象的影响.结果表明:在pH4～11范围内ANS的荧光寿命呈U字形分布;其荧光寿命中含有快慢两种衰减成分;随着介质pH的变化;这两种成分的含量所占比例也有很大变化.说明介质pH能够引起蛋白构象的变化.
By measuring fluorescent lifetime of AXS binding with purple membrane, the pH - dependence of bacteriorhodopsin conformation was studied. Results show that with the merease of pH. the lifetime decreases in actcie mediurn and increases in basic medium.The biletime contains fast and slow components, and the percentage of two component, changes with the change of pH, The results show that pH of medium can cause the change of protein conformation.
