摘要
以钝齿棒杆菌102S_2-58为出发菌株,在原生质体彤成及再生的最佳条件下制备原生质体,并对原生质体进行紫外诱变处理,对大量的再生突变株进行发酵筛选,获得了高产稳定株102-100号,其发酵液经氨基酸自动分析仪测定L-赖氨酸积累量由出发菌株的50.0mg/ml提高到80.8mg/ml,糖转化率达到63.88%。发酵液中主要副产酸——缬氨酸和蛋氨酸的量明显降低。
Under the optimun conditions of formation and regeneration of protoplasts, protoplasts of Corynebacterium crenatum 102S_2-58 were prepared. The highyield stable strain 102-100 were obtained by UV irradiation of protoplasts and screening of the fermentation of many regenerative mutants. Amino acids in the fermentation broth of the strain 102-100 were determined by the automatic amino acid analyzer. The strain 102-100 produced large amounts of L-lysine (80.8mg/ml) than parental strain 102S_2-58 which produced 50.0mg/ml of Llysine. The conversion rate of glucose was 63.88%. The yields of other amino acids, such as valine and methionine, were reduced obviously.
