摘要
真养产碱菌(Alcaligenes eutrophus)H16(ATCC 17699)能以丁酸为唯一碳源在无氮合成培养基中积累大量生物可降解的聚β-羟基丁酸(PHB)。将此菌在丰富培养基中预培养36小时后转入无氮合成培养基甲,在30℃、pH8.0条件下培养48小时,积累的 PHB 可达细胞干重的63%(w/w)。较高浓度的丁酸对 PHB 的积累有抑制作用。采用丁酸自动流加法控制 pH 值的工艺可降低丁酸的抑制作用。此时 PHB 的产率达0.4g/g 丁酸,PHB 比产生率(ρ)达0.1g·g 生物量^(-1)·h^(-1)。
Bacteria Alcaligenes eutrophus H16(ATCC 17699)can utilize butyric acid as the sole car-bon source to produce poly-β-hydroxybutyric acid(PHB)in nitrogenfree medium.The opti-mal conditions for PHB production were:36 h of first stage cultivation;temperature,30℃;pH 8.0;48 to 72 h of the second state growth.The maximum content of PHB obtained in batchculture was 63%(w/w).There was the inhibition of butyric acid to PHB accumulation inbatch culture.The fed-batch culture was proceeded by feeding butyric acid to keep pH at con-stant.The yield of PHB production was about 0.4g PHB/g butyric acid,and the specific pro-duction rate of PHB was about 0.1g PHB·g biomass^(-1)·h^(-1).
