摘要
以微胶囊法固定平菇半纤维素酶,探讨固定化条件与固定化酶的酶学性质。结果表明:当海藻酸钠浓度为3%,戊二醛浓度为1%时,固定化酶有较高的活力回收率;固定化酶的最适温度比游离酶提高了10℃,最适pH值较游离酶向酸性范围移动0.4个单位;游离酶和固定化酶的米氏常数分别为0.906 g/L和0.937 g/L;固定化酶的耐热性较好,在60~80℃下仍能保持较高的酶活力,具有良好的贮存稳定性。常见的大部分金属离子对固定化酶的活力影响较小。
Hemicellulase from Pleurotus ostreatu was immobilized by microcapsule methods.The immobilization conditions and the characteristics of immobilized hemicellulase were investigated.The results showed that the recovery of immobilized hemicellulase activity was high when the concentrations of sodium alginate and glutaraldehyde were 3.0% and 1.0%,respectively.In comparision with free enzyme,the optimum reaction temperature for immobilized hemicellulase was increased by 10 ℃ and the optimal pH of the immobilized hemicellulase shifted 0.4 to acidity.The Km values of free enzyme and the immobilized hemicellulase were 0.906 g/L and 0.937 g/L,respectively.The thermal stability of the immobilized hemicellulase was greatly improved,maintaining higher enzyme activity at 60-80 ℃.The effects of common metal ions were insignificant on the immobilized hemicellulase activity.
