摘要
目的:本研究通过观察受体型蛋白酪氨酸磷酸酶(PTPRJ)在大鼠肝星状细胞中的表达及姜黄素对其的影响,探讨以PTPRJ为靶点,姜黄素对抗肝纤维化的分子机制。方法:四氯化碳(CCL4)皮下注射诱导大鼠肝纤维化模型,并采用Percoll梯度离心法分离培养原代大鼠肝星状细胞,采用western-blot、RT-PCR法分别测定PTPRJ、其mRNA在肝组织及肝星状细胞的表达变化及姜黄素对其表达的影响。结果:western-blot、RT-PCR检测结果显示:肝组织和肝星状细胞中PTPRJ表达呈阳性,并随着肝纤维化进展,其表达逐渐降低;姜黄素作用后,肝组织和肝星状细胞PTPRJ表达明显增多。结论:姜黄素可显著增强肝星状细胞PTPRJ的表达,从而改善或逆转肝纤维化进展。
Objective:Protein tyrosine phosphatase,receptor-type J(PTPRJ)is expressed in a wide variety of tissues including liver.The aim of this study was to investigate the expression of PTPRJ in rat hepatic stellate cell(HSC)and the influence of curcumin on this.Methods:In CCL4,intoxicated rat livers and HSCs,PTPRJ expression was measured by western-blot and RT-PCR.And then we observed the influence of cmrcumin on PTPRJ' expression.Results:In CCl4-intoxicated rat livers,PTPRJ expression was decreased with time,as the IeveI of hepatic fibrosis increased.But PTPRJ expression observed by western-blot and RT-PCR was gradually stronger after curcumin treated.Conclusion:PTPRJ was expressed in rat HSCs.It may have negative effect on HSC activation,proliferation and liver fibrosis.And curcumin treated can be stronger its expression.
