摘要
目的:受体型蛋白酪氨酸磷酸酶(PTPRJ)在多种组织包括肝内表达。目前认为其作为一种潜在抑癌基因对于细胞增殖及迁徙具有抑制作用。文章通过观察PTPRJ在大鼠肝星状细胞中的表达及功能,探讨PTPRJ在肝纤维化发生中的意义。方法:四氯化碳(CCl4)皮下注射诱导大鼠肝纤维化模型,用Western技术检测PTPRJ在肝纤维化组织中的表达变化,采用Percoll梯度离心法分离培养原代大鼠肝星状细胞,采用细胞免疫荧光法观察PTPRJ表达情况,RT-PCR法测定其mRNA在肝星状细胞体外活化过程中的表达变化。结果:免疫荧光及RT-PCR显示肝星状细胞中PTPRJ表达呈阳性;在CCl4诱导的实验性大鼠肝纤维化模型中随着不同时间段纤维化程度加重,PTPRJ表达减少;原代大鼠肝星状细胞体外培养自活化过程中,RT-PCR测定的PTPRJ表达逐步减少。结论:PTPRJ在原代肝星状细胞中表达为阳性,并在肝星状细胞活化及肝纤维化过程中表达减少,提示PTPRJ可能对于肝纤维化发生发展起负性调控作用。
Objective:Protein tyrosine phosphatase, receptor-type J(PTPRJ) is expressed in a wide variety of tissues including liver. A growing body of evidence implicates PTPRJ as a potential anti-oncogene in the inhibition of cell growth and cell migration. The aim of this study was to investigate the expression and functions of PTPRJ in rat hepatic stellate cell (HSC) and reveal the function of PTPRJ in hepatic fibrosis. Methods :In CC14-intoxicated rat livers, PTPRJ expression was measured by West-blotting. In isolated rat HSCs and hepatic stellate cell lines:HSC-T6, PTPRJ expression was examined by immunofluorescence and reverse transcriptase polymerase chain reaction. Results:In CC14-intoxicated rat livers, PTPRJ expression was decreased with time, as the level of hepatic fibrosis increased. PTPRJ immunoreactivity was found in primary rat HSCs. PTPRJ mRNA expression was gradually reduced in the self-activation process of HSCs in vitro, with the extension of culture time. Conclusion :PTPRJ was expressed in rat HSCs; and PTPRJ expression decreased during liver fibrosis process in vivo and during cell culture course with HSC' s activation in vitro. PTPRJ may have negative effect on HSC activation, proliferation and liver fibrosis.
