摘要 目的:研究肿瘤转移相关基因1(MTA1)与肿瘤转移抑制基因KISS-1蛋白在胃癌的表达及临床意义。方法:采用免疫组化的方法检测20例胃正常组织和82例胃癌组织中MTA1和KISS-1的表达。结果:MTA1与KISS-1蛋白在胃癌组织中阳性表达率分别为52.44%和40.24%,在正常胃组织的阳性表达率分别为5.00%和90.00%;MTA1蛋白高表达与胃癌浸润深度、组织学分级、临床分期、淋巴结转移和远隔转移的发生比较差异有统计学意义(P<0.01);KISS-1蛋白低表达与胃癌浸润深度、临床分期、淋巴结转移和远隔转移的发生比较差异有统计学意义(P<0.05);胃癌组织中MTA1与KISS-1蛋白表达呈负相关(γs=-0.364,P<0.01)。结论:MTA1蛋白水平高表达和KISS-1蛋白水平低表达与胃癌的浸润、转移密切相关,可作为诊断胃癌患者转移、复发和判断预后的参考指标,有望成为胃癌基因治疗新的靶点。 Objective:To investigate the expression and clinical significance of metastasis associated gene 1(MTA1) tumor metastasis suppressor gene KISS-1 in human gastric carcinoma. Methods:Immunnohistochemical technique was used to detect the MTAl and KISS-1 protein expression in 20 normal human gastric mucosa and 82 human gastric carcinoma samples.Results:The positive rate of MTA1 and KISS-1 protein was respectively 52.44% and 40.24% in gastric carcinoma and 5.00% and 90.00%in the normal gastric mucosa. Over-expression of MTA1 was significantly involved in the depth of invasion,grade,clinical stage,distant metastasis and lymph node metastasis(P0.01),and the similar results were found with the low expressed KISS-1 except the grade(P0.05).Yet,MTA1 expression was negatively related to KISS-1 in human gastric carcinoma (P0.01). Conclusion:Over-expression of MTA1 and low KISS-1 expression were significantly involved in infiltration and metastasis of the human gastric carcinoma,and the findings suggest that MTA1 and KISS-1 may be useful markers for evaluation of tumor malignancy,predication of recurrence,metastasis and prognosis in gastric carcinoma patients,which is likely to be new target of gene therapy of gastric carcinoma.
