针药结合对急性局灶性脑缺血-再灌注大鼠脑组织自由基代谢的影响 被引量:8

摘要 目的:观察针药结合治疗对局灶性脑缺血-再灌注大鼠模型脑组织SOD活性、MDA及NO含量的影响,探讨针药结合治疗对缺血性脑血管病病理状态的干预机制。方法:120只大鼠随机分为两组,分别于缺血2 h再灌注后1 h和再灌注后13 h进行治疗,并分别于再灌注后12 h(I2 h/R12 h组)和再灌注后1 d(I2 h/R1 d组)断头开颅取脑,检测脑组织SOD活性及MDA和NO含量。每组又分为4个亚组,分别是模型组、假手术对照组、头针组及针药组。除假手术对照组外,各组大鼠均采用线栓法建立局灶性脑缺血-再灌注模型,头针组以"百会"透"曲鬓"治疗,针药组以补阳还五汤灌胃结合"百会"透"曲鬓"治疗。结果:①在I2 h/R12 h组里,模型组大鼠脑组织SOD活性明显下降,MDA和NO含量显著升高;头针组和针药组大鼠脑组织SOD活性升高,MDA和NO含量降低。②在I2 h/R1 d组里,针药组MDA含量显著低于头针组。结论:对于脑缺血-再灌注模型鼠,头针及针药结合治疗可抑制自由基代谢及改善线粒体功能,减轻氧自由基介导的脂质过氧化反应。 Objective:To observe the effect of electric acupuncture combined with herbs on SOD activity and contents of MDA and NO in brain tissue of acute focal cerebral ischemia reperfusion model rats.Methods:120 rats were randomly di-vided into 2 groups(I2 h/R12 h group and I2 h/R1 d group),then each group was divided into 4 groups,model group,sham operation group,electric acupuncture group,electric acupuncture combined with herbs group.Model rats were con-structed by suture method.The electric acupuncture group rats were punctured at Baihui point through Qubin point.SOD activity,contents of MDA and NO in brain tissue of rats were measured.Results: ①In I2 h/R12 h group,compared with sham operation group,SOD activity was decreased(P0.01),contents of MDA and NO were increased significantly in mod-el subset(P0.05);compared with model group,SOD activity was increased(P0.05),contents of MDA and NO were de-creased in electric acupuncture group and electric acupuncture combined with herbs group.②In I2 h/R1 d group,MDA content in electric acupuncture combined with herbs group was significantly lower than that of electric acupuncture group.Conclusion:Traditional Chinese medicine therapy can restrain the production of free radicals,better the function of mito-chondria,alleviate lipid peroxidation induced by free radicals.
