电针干预对老龄大鼠脑缺血再灌注海马内源性神经干细胞增殖分化的影响 被引量:11

Effect of electroacupuncture on cell proliferation and differentiation in dentate gyrus of aged rats with focal cerebral ischemia reperfusion injury

摘要 目的:从海马齿状回(dentate gyrus,DG)细胞增殖和分化的变化揭示针刺穴位抗老年脑缺血再灌注损伤的保护机制。方法:采用MCAO方法复制脑缺血动物模型,免疫组化方法检测观察DG的5-溴脱氧尿嘧啶(Brdu)、神经元核抗原(NeuN)及胶质纤维酸性蛋白(GFAP)阳性细胞数,观察脑缺血3h,I/R1、3、7、14、21d大鼠神经症状积分、脑组织含水量、病理变化、海马齿状回细胞增殖、分化的变化。结果:模型组脑组织含水量(I/R1、3、7d)、神经症状积分(各时间点)、Brdu阳性细胞(各时间点)、BrdU/NeuN双标阳性细胞(各时间点)BrdU/GFAP双标阳性细胞(I/R7、14、21d)数目均高于假手术组(P<0.01),各指标和非穴位组无显著性差异;穴位治疗组神经症状积分、脑组织含水量、(I/R3、7、14d)低于模型组和非穴位治疗组(P<0.05或P<0.01),Brdu阳性细胞(各时间点)、BrdU/NeuN双标阳性细胞数目(各时间点)高于模型组和非穴位组(P<0.05,P<0.01),BrdU/GFAP双标阳性细胞(I/R3、7、14d)变化相反(P<0.05,P<0.01)。结论:脑缺血可激活大鼠DG神经干细胞增殖;针刺穴位抗脑缺血再灌注损伤的机制与其促进DG区神经干细胞增殖和分化有关。 Objective:To study the protective effect of electroacupuncture on stem cell proliferation and differentiation in dentate gyrus (DG) in aged rats with focal cerebral ischemia reperfusion injury. Methods:We duplicated focal cerebral ischemia model with MCAO,with ischemia 3h,I/R1,3,7,14,21d points. The effect of electroacupuncture on the nervous dysfunction score,the water content of cerebral constitution and the stem cell proliferation and differentiation in dentate gyrus were observed,and the non-acupoint group was as control. Results:The nervous dysfunction score (all the time points),the water content of cerebral tissue (I/R1,3,7d),BrdU positive cells (all the time points),BrdU/NeuN positive cells (all the time points),BrdU/GFAP positive cells (I/R7,14,21d),in model group were higher than those of the sham-operated group (P0.01),there was no difference of model group from the nonacupoint group. The nervous dysfunction score,the water content of cerebral tissue (I/R3,7,14d),BrdU/NeuN positive cells(I/R3,7,14d)in electroacupuncture group were lower than those in model group and non-acupoint group respectively (P0.05,P0.01),it was contrary to BrdU/GFAP positive cells(I/R7,14,21d)(P0.05,P0.01). Conclusion:Neural stem cells in DG were activated after cerebral ischemia. Electroacupuncture could protect brain cell from damage after focal cerebral ischemia reperfusion injury by promoting cell proliferation and differentiation in DG in aged rats.
