酪素季铵盐衍生物的合成、表征与抗菌性能 被引量:4

SYNTHESIS,CHARACTERIZATION AND ANTIBACTERIAL PROPERTY OF QUATERNARY AMMONIUM SALT DERIVATIVES OF CASEIN

摘要 以硝酸铈铵为引发剂,以甲基丙烯酰氧乙基-十八烷基-二甲基溴化铵为活性单体,与酪素进行接枝共聚反应制备季铵盐型酪素衍生物.用核磁共振、红外光谱、热重分析仪与激光粒度Zeta电位仪对接枝产物进行了表征和测定,用悬菌定量杀菌试验研究了其抗菌性能.结果表明:甲基丙烯酰氧乙基-十八烷基-二甲基溴化铵季铵盐单体通过其双键接枝到酪素表面;酪素的零电位pH=4.6,而酪素季铵盐衍生物的零电位pH=10.4,由于阳离子季铵基团的引入使酪素的零电位发生了较大的位移;所制备的酪素季铵盐衍生物对金黄色葡萄球菌、大肠杆菌与白色念珠菌在振荡作用15 m in后,平均杀菌率分别为99.99%、99.99%和92.33%. The quaternary ammonium salt derivatives of casein were prepared by graft copolymerization of methacryloxylethyl-octadecyl-dimethyl ammonium bromide monomer and casein in the presence of ceric ammonium nitrate as the initiator.The graft product was characterized and determined by hydrogen nuclear magnetic resonance spectroscopy(1H NMR),Fourier transform infrared spectroscopy (FTIR),thermogravimetric analyzer(TGA),and laser particle size and zeta potential analyzer,and the antibacterial properties of the product were tested by the suspension quantitative test.The results show that methacryloxylethyl-octadecyl-dimethyl ammonium bromide monomer is grafted onto the surface of casein through double bonds;the zero potential pH value of casein is equal to 4.6,and the zero potential pH value of quaternary ammonium salt derivatives of casein is equal to 10.4.Accordingly,the zero potential of casein has large shift after the introduction of cationic quaternary ammonium group.The produced quaternary ammonium salt derivatives of casein had high effects in inhibiting E.coil(8099),S. aureus(ATCC 6538) and C.albicans(ATCC10231) after shaking for 15 min,and the average sterilization rates were 99.99%,99.99% and 92.33%,respectively.
