威草胶囊对尿酸性肾病大鼠肾组织结缔组织生长因子和肝细胞生长因子的影响 被引量:8

Effects of Weicao Capsules on Expression Levels of Connective Tissue Growth Factor and Hepatocyte Growth Factor in Renal Tissues of Uric Acid Nephropathy Rats

摘要 目的观察威草胶囊对尿酸性肾病大鼠肾组织中结缔组织生长因子(connective tissue growth fac-tor,CTGF)和肝细胞生长因子(hepatocyte growth factor,HGF)表达的影响,探讨其对尿酸性肾病的治疗作用及其机制。方法将50只SD雄性大鼠随机分为正常对照组、模型组、别嘌呤醇组、威草胶囊小剂量组和威草胶囊大剂量组5组。用酵母和腺嘌呤合用进行模型复制,模型复制成功后分别给予9.0 g/L氯化钠注射液、别嘌呤醇和威草胶囊灌胃。治疗前及治疗4周末,分别检测各组大鼠血中尿素氮(blood urea nitro-gen,BUN)、肌酐(creatinine,Cr)、尿酸(uric acid,UA)水平,采用免疫组织化学技术检测大鼠肾组织CTGF和HGF的表达情况。采用苏木精-伊红染色观察各组大鼠肾组织的病理变化。结果与正常对照组比较,模型组血UA、BUN、Cr均明显升高(P<0.01),模型组肾组织中CTGF表达水平明显升高,HGF表达水平明显降低;与模型组比较,威草胶囊大、小剂量组和别嘌呤醇组大鼠血UA、BUN、Cr水平及肾组织中CTGF的表达水平明显降低(P<0.01),威草胶囊大、小剂量组和别嘌呤醇组肾组织中HGF的表达水平明显升高(P<0.01);威草胶囊大、小剂量组和别嘌呤醇组之间上述指标比较,差异均无显著性(P>0.05)。病理检测显示,大、小剂量威草胶囊和别嘌呤醇可减轻肾间质尿酸盐结晶沉积和炎性细胞浸润,以及肾小管纤维化程度。结论威草胶囊能够改善尿酸性肾病大鼠模型的肾功能,其机制可能与其降低血尿酸,减少尿酸盐结晶在肾小管的沉积,以及减少CTGF和增加HGF在尿酸性肾病模型大鼠肾组织中的表达,阻止肾间质纤维化的发展等有关。 Objective To investigate the effects of Weicao Capsules, a traditional Chinese medicine mainly containing Radix Clematidis, Semen Cassiae, etc. , on the expression levels of connective tissue growth factor (CTGF), and hepatocyte growth factor (HGF) in renal tissues of uric acid nephropathy rats, and to explore its therapeutic effects and its mechanism of action. Methods Fifty SD male rats were randomly divided into 5 groups: normal control group, model group, allopurinol treated group, low dosage Weicao Capsules treated group and high dosage Weicao Capsules treated group. The rat model of uricacid nephropathy was induced by feeding yeast 10 g/kg and adenine 100 mg/kg during 19 days, successively. Rats in normal control group were given normal saline; and rats in allopurinol treated group, and low- and high- dosage Weieao Capsules treated groups were treated with intragastric injection of corresponding drugs for four weeks. Before treatment and after four weeks of treatment, serum levels of blood urea nitrogen (BUN), creatinine (Cr), uric acid (UA) in each group were determined, respectively; and immunohisto- chemistry method was used for detecting the expression levels of CTGF and HGF in renal tissues of uric acid nephropathy rats. Pathological changes of renal tissues of rats in all groups were observed using hematoxylin-eosin staining. Results In comparison with those of normal control group, serum levels of BUN, Cr, and UA in model group were significantly increased (P〈0.01) ; the expression level of CTGF was significantly Increased (P〈0. 0]), and the expression level of HGF was significantly deereased(P〈0.01)in renal tissues of rats in model group. In comparison with those of the model group, the serum levels of BUN,Cr, and UA as well as the expression level of CTGF were significantly decreased (P〈0.01), and the expression level of HGF was significantly increased in lowand high-dosage Weicao Capsules trea- ted groups and allopurinol treated group (P〈0.
