摘要
目的观察威柏膏外敷治疗对急性痛风性关节炎模型大鼠关节滑膜组织炎性反应的调控作用。方法雄性Wistar大鼠随机分为正常对照组、模型对照组、威柏膏组、秋水仙碱组。观察大鼠步态及受试踝关节肿胀度,HE染色观察滑膜组织病理改变,免疫组化法检测踝关节滑膜组织中NALP-3炎性体的核心蛋白NALP-3及效应分子Caspase-1的相对表达含量。结果与模型对照组比较,威柏膏组可显著减少炎性细胞浸润,并降低受试关节滑膜组织中NALP-3、Caspase-1蛋白的相对表达含量(P<0.05)。步态评分、关节肿胀度指标方面,威柏膏组及秋水仙碱组均较模型对照组明显改善,且威柏膏组改善更显著。结论威柏膏外敷能减轻急性痛风性关节炎模型大鼠的关节炎性反应,其机制可能是下调了滑膜组织中NALP-3炎性体的表达水平。
Objective:To observe the regulatory effect of Weibo Ointment on inflammatory response of synovial tissue in rats with acute gouty arthritis.Methods:Male Wistar rats were randomly divided into normal control group,model control group,Weibo Ointment group and colchicine group.The gait of rats and the swelling degree of ankle joints were observed.The pathological changes of synovium were observed by HE staining.The relative expression levels of NALP-3 and Caspase-1 in synovial tissue of ankle joint were detected by immunohistochemistry.Results:Compared with model control group,Weibo Ointment group could significantly reduce inflammatory cell infiltration and the relative expression of NALP-3 and Caspase-1 protein in synovial tissue(P < 0.05).Conclusion:Weibo Ointment can alleviate the inflammatory reaction of acute gouty arthritis model rats,and its mechanism may be related to down-regulating the expression of NALP-3 inflammatory body in synovial tissue.
