Some Biological Characteristics of Avian Escherichia coli

摘要 对鸡源大肠埃希氏菌的某些生物学特性进行了研究,包括对1日龄鸡的致病性、溶血性、菌毛的表达、血凝性、对鸡胚成纤维细胞(CEF)的粘附特性、体内外与鸡气管粘膜的粘附特征、质粒特征、全菌蛋白SDS-PAGE电泳及免疫转印等。结果表明,鸡源大肠埃希氏菌的溶血性、质粒特征、对CEF的粘附特性与其致病性无关;菌毛与致病性有关,大部分菌株表达Ⅰ型菌毛;血凝能被D-甘露糖抑制,不同菌株血凝谱具有差异;体内外与气管粘膜的粘附特性与菌株致病性有关;在全菌蛋白SDS-PAGE电泳中,相对分子质量约为40700的多肽仅存在于具有致病性的菌株中,经免疫转印证实。 Some strains of avian E. coli were studied with respect to their some biological characteristics, including pathogenicity for 1 day old chick, hemolysin production, hemagglutination adhesion to CEF cells, plasmid DNA profile and SDS PAGE electrophoresis of whole bacterial proteins, electron microscopic observation for fimbriae expression, as well as adhere to chick tracheal mucosa in vitro and in vivo. The results showed that a polypeptide of 40 700 exists in and is specific for the pathogenic strain of E. coli and that the hemolysin production, plasmid profile and adhesion to CEF cells were not related to the pathogenicity of these strains. Fimbriae type Ⅰ expressed by majority of these strains was correlated to their pathogenicity. 1 (type Ⅰ) fimbriae, and exhibited mannose sensitive hemagglutination (MSHA), the scope of HA were different among these strains. Adhere to tracheal mucosa in vitro and in vivo were related to their pathogenicity. SDS PAGE of whole bacterial proteins of different strain which had no pathogenicity lacked one major protein subunits of 40 700 found only in pathogenic strains for 1 day old chick, western blotting revealed that this major protein subunit exist only in pathogenic avian E. coli isolates.
