线栓法制备大鼠局灶性脑缺血再灌注模型的改进与体会 被引量:6

摘要 目的 改进线栓法制作动物大脑中动脉缺血再灌注模型,并通过测定多个指标评价该方法的价值.方法 采用体重250~300g雌雄各半的Wistar大鼠,分成改良线栓法组、传统线栓法组与假手术组.术后评价的指标包括手术成功率和手术时间、大鼠神经功能缺损评分、大鼠体重、TTC染色、HE染色.结果 改良线栓法组手术成功率高于传统线栓法组,手术时间短于后者;前者神经功能缺损评分与后者比较,差异无显著性;TTC染色可见明显脑缺血部位;HE染色可见梗塞侧脑细胞有大片坏死和凋亡,细胞稀疏.结论 改良线栓法简便可靠,模型成功率和稳定性较好,是较理想的MCAO(middle cerebral artery occlusion)模型制作方法. Objective To modify the method of preparing focal cerebral ischemia-reperfusion model in rats and evaluate its application according to several indexes. Methods Male and female Wistar rats of 250 - 300g were divided into improved suture group, traditional suture group and sham operated group. The post-operative evaluation indexes include achievement ratio and time,rats'neurologic impairment score,TTC dyeing,HE dyeing. Results Both the operation achievement ratio and operation time of the improved suture group were better than traditional ones. There was no significant statistical difference between improved and traditional suture group in rats'neurologic impairment score, TTC dyeing showed obvious location of ischemia. HE dyeing showed a large sheet of cellular necrosis and apoptosis,sparse cell distribution in infarction side of the brain. Conclusion The modified method in preparation of rat model of local cerebral ischemia-reperfusion is convenient and safe; the model achievement ratio and stability are well;this method is ideal for MCAO( middle cerebral artery occlusion) model huihting.
