人参总皂苷对大鼠脑缺血再灌注损伤后神经血管单元的保护作用 被引量:4

摘要 目的研究人参总皂苷对缺血再灌注大鼠脑组织高迁移率族1(HMGB1)表达的影响,探讨人参总皂苷对大鼠脑缺血再灌注损伤后的神经血管单元的保护作用。方法 96只雄性SD大鼠随机分为正常组、模型组和高、低剂量治疗组,正常组只暴露一侧颈总动脉后缝合,不予任何处理;模型组和人参总皂苷高、低剂量治疗组应用改良线栓法制备成功后立刻分别应用相应药物腹腔注射,在1d,3 d,5d,7d时间点分别取脑组织,通过HE染色观察神经损伤程度,免疫组化的方法检测HMGB1的表达变化。结果人参总皂苷高、低剂量组HMGB1蛋白表达较模型组明显减少(P均<0.05),人参总皂苷高、低剂量组HMGB1蛋白表达比较有显著性差异(P均<0.05)。结论人参总皂苷通过抑制HMGB1/RAGE/ICAM-1途径缓解缺血再灌注造成的血脑屏障的损害,在一定范围内抑制HMGB1的表达,随剂量的加大抑制幅度增加,从而降低炎症反应,抑制脑缺血再灌注周围神经细胞凋亡,促进细胞代谢,对脑缺血再灌注大鼠大脑起保护作用。 Objective To discuss the iniluence of Saponins of panax ginseng on HMGB1 and its protective effect on neurocascular unit. Methods The rats were randomly divided into quadrisection:normol,model and high,low dosage of Saponins of panax ginseng groups, the nonnol group was keep intact but worked with line embolism to block the middlecerebralartery. Model group,high and low dosage of Saponins of panax ginseng groups were remedied by the medi- c.inc. The brain slices were gotten at l d,3d,5d,7d to oberserve the change of HMGB1 by immunohistochemistry. Results Compared the handled groups to the normol group, HMGBlprotein expression was obvious decl^ased in the surgel7 group. The expression of HMGBI protein was obviously decreased in high anti low dosage of Saponins of panax ginseng groups,compared with the model group( P 〈 0.05). The distinct of the expression of HMGBI between high and low dose of Saponins of panax ginseng groups was valuable(P 〈 0.05 ). Conclusion Saponins of panax ginseng has pl^tty good protection on cerebral protection in the rat with cerebral ischemia reperfusion throw decreasing the way of HMGBI/ RAGE/ICAM-I, and the more dose,the more effectived.
