摘要 目的研究阿魏胶囊对载脂蛋白A4(Apo-A4)基因转录水平的影响。方法将大鼠随机分为正常组、模型组和阿魏胶囊预防中、高剂量组及阿魏胶囊治疗中、高剂量组,共6个组。除正常组仅用普通饲料喂养外,其余各组每天灌以高脂乳剂2mL。其中预防2组造模当天灌服阿魏胶囊药物混悬剂4周;治疗2组连续造模4周后,待造模成功,再灌服阿魏胶囊药物混悬剂4周。各组取血液后采用逆转录-聚合酶链反应(RT-PCR)对Apo-A4进行检测,观察其表达水平。结果RT-PCR凝胶成像系统结果分析表明,大鼠Apo-A4基因的转录水平正常组与模型组相比,差异有统计学意义(P<0.05);模型组与预防中、高剂量组及治疗中、高剂量组相比差异有统计学意义(P<0.01)。治疗中剂量组和预防中剂量组的转录水平都略高于治疗高剂量组和预防高剂量组。结论阿魏胶囊明显地提高或稳定Apo-A4基因的转录水平。 Objective To study the influence of Awei Capsules on level of apoprotein A4(Apo-A4) genetic transcription.Methods All rats were randomly divided into the normal control group,model group,prevention groups with mid-dose and high-dose Awei Capsules and treatment groups with mid-dose and high-dose Awei Capsules.The normal control group was fed normally and other groups were given orally high fat emulsion (2 mL).The two prevention groups were given the suspension of Awei Capsules for 4 weeks beginning from the day of modeling,and the two treatment groups were modeled continuously for 4 weeks until success,and then given the suspension for 4 weeks.The level of Apo-A4 genetic transcription in all groups was detected by applying reverse transcription-polymerase chain reaction (RT-PCR).Results The outcomes of RT-PCR showed that there was a significant difference in the level of Apo-A4 genetic transcription between the normal control group and model group (P0.05).There was a significant difference between the model group and prevention groups with mid-dose and high-dose Awei Capsules or treatment groups with mid-dose and high-dose Awei Capsules (P0.01),respectively.Conclusion Awei Capsules can significantly increase or stabilize the level of Apo-A4 genetic transcription.
