摘要
目的:探讨海马神经元核受体-盐皮质激素受体(MR)参与下丘脑-垂体-肾上腺(HPA)轴调节的机制.方法:建立大鼠穹窿切断模型,采用ELISA方法检测穹窿切断0、4、7、10d后血中糖皮质激素的浓度,建立大鼠穹窿切断同时摘除肾上腺7d的模型,采用免疫组织化学、免疫印迹方法检测海马MR表达的变化.结果:穹窿切断7d后血中糖皮质激素的浓度升高,穹窿切断同时摘除肾上腺7d后,海马神经元MR表达升高.结论:海马核受体MR的表达变化可能与糖皮质激素的浓度有关,且为负相关.
Objective: To explore the mechanisms of mineralocorticoid receptor (MR) which is one of nuclear receptors in the hippocampal participation in regulating HPA axis. Methods: The fornix was transeeted to set an animal model, and ELISA was used to detect glucocorticoid concentrations on day 0,4,7 and 10 after fornix transection. Immunohistoehemical method and Western blotting were respectively used for observation and quantitative detection of the changes of MR expression in the hippoeampal neurons after fornix transection and adrenalectomy. Results: Glucocortieoid concentrations was increased on day 7 after fornix transection. The expression of MR was up-regulated on day 7 after fornix transection and adrenalectomy. Conclusion: The expression of rnineralocorticoid receptor which is one of neuclear receptors in the hippocampal neurons is negatively associated with the glucocorticoid concentrations.
