摘要
目的:观察下丘脑促肾上腺皮质激素释放激素(corticotropin releasing hormone,CRH)与糖皮质激素的关系,探讨海马参与HPA轴调节的机制。方法:建立大鼠穹窿切断模型,采用ELISA方法检测穹窿切断0、4、7、10 d血中糖皮质激素的浓度;建立大鼠穹窿切断同时摘除肾上腺7 d的模型,采用免疫组织化学、免疫印迹方法检测CRH的表达变化。结果:穹窿切断7 d后血中糖皮质激素的浓度升高,穹窿切断同时摘除肾上腺7 d后,下丘脑CRH表达升高。结论:海马通过穹窿对HPA轴发挥负反馈调节作用,糖皮质激素负反馈调节下丘脑CRH。
Objective: To observe the relation between corticotropin releasing hormone(CRH) and glucocorticoid and to explore the mechanism of the hippocampus involved in HPA axis regulation.Methods: The fornix was transected to establish a animal model,ELISA was used for detection of glucocorticoid concentrations on day 0,4,7 and 10 after fornix transaction.Immunohistochemical method and Western Blotting were respectively used for the expression of CRH in the hypothalamus after fornix transection and adrenalectomy.Results: Glucocorticoid concentration was increased on the 7th day after fornix transection.The expression of CRH was up-regulated on the 7th day after fornix transection and adrenalectomyon.Conclusions: The hippocampus provides a negative feedback to the HPA axis through fornix,the glucocorticoid has a negative feedback effect on CRH.
