美味牛肝菌胞外多糖的分离纯化及组分分析 被引量:4

摘要 主要对美味牛肝菌发酵产生的胞外多糖(BeBEP)进行了分离纯化和组分的分析。方法:采用DEAE-纤维素柱层析和SephadexG-100凝胶柱层析对BeBEP进行分离纯化,得到的主要成分BeBEPⅢa,用紫外光谱和醋酸纤维素薄膜电泳方法进行纯度鉴定,红外光谱进行结构分析,高效液相色谱法分析单糖组分。结果表明:BeBEPⅢa为均一组分,存在吡喃糖苷,由D-木糖、D-甘露糖和D-半乳糖三种单糖组成,摩尔比为3.1∶63.3∶1.0,其中主要以D-甘露糖为主。 This article focused on the separation and purification and analysis of the fermentation components of BeBEP. The BeBEP was separated and purificated by DEAE-cellulose and Sephadex G-100 column chromatography,the purity of which was verified through ultraviolet spectrum and cellulose acetate pellicle electrophoresis,the structure of which was analyzed with IR spectrum and the monosaccharide component was analyses with HPLC. The results showed that BeBEPⅢa was a polysaccharide,presences pyran indican,including xylose,mannose and galactose with a molar ratio of 3.1∶63.3∶1.0,but mannose was primary.
