哮喘和免疫耐受大鼠淋巴液中Treg变化及其功能状态的实验研究 被引量:2

The experimental study on the change and function of Treg cells from lymph in bronchial asthmatic and the immunologic tolerance rat

摘要 观察CD4^+CD25^+Foxp3^+调节性T细胞、IL-10、TGF-β在哮喘和免疫耐受大鼠淋巴液与血液中的水平,探讨其与哮喘及免疫耐受的关系。建立大鼠哮喘和免疫耐受模型,收集激发后0 h、24 h、48 h淋巴液、血液中淋巴细胞,采用流式细胞术检测淋巴液及血液中CD4^+CD25^+Foxp3^+T细胞百分率;采用ELISA方法检测血浆及淋巴液中IL-10、TGF-β水平。结果显示,哮喘组淋巴液和血液的CD4^+CD25^+Foxp3^+T细胞百分率水平、血浆IL-10和TGF-β浓度在各时间点均低于对照组和耐受组(P<0.05);哮喘组和耐受组淋巴液中CD4^+CD25^+Foxp3^+T细胞百分率水平、IL-10浓度在不同时间点均高于血液水平(P<0.05),而TGF-β浓度则低于血液水平(P<0.05);耐受组淋巴液和血液中CD4^+CD25^+Foxp3^+T细胞百分率水平、血浆IL-10和TGF-β浓度与对照组相比无显著性差异(P>0.05)。说明哮喘大鼠淋巴液中存在CD4^+CD25^+Foxp3^+调节性T细胞数量以及功能失调,且淋巴液中CD4^+CD25^+Foxp3^+调节性T细胞水平显著高于其血液水平,而建立免疫耐受后大鼠淋巴液及血液中CD4^+CD25^+Foxp3^+调节性T细胞数量升高,说明通过免疫耐受可以避免哮喘发病或减轻症状,推测通过建立免疫耐受可能成为一种治疗哮喘的有效途径。 To observe the expression of CD4+CD25+Foxp3+ Tregs, IL-10 and TGF-β from lymph and blood in bronchial asthmatic and immunologic tolerance rat, and to investigate the relationship between asthma and immunologic tolerance, lymph and blood samples of 0 hour. 24 hour and 48 hour after the last challenge were collected from the rat model of asthma and immunologic tolerance, and the percentage of CD4+CD25+Foxp3+ Treg were detected by flow cytometery(FCM), while the levels of II. 10 and TGF-β were determined by ELISA. It was found that the percentage of CD4+CD25+Foxp3+ Treg in the blood and the levels of IL 10 and TGF-β in the plasma in cases of asthmatic group were significantly lower than that of the control and tolerance group at different times(P〈0.05). The percentage of CD4+ CD25+ Foxp3+ Treg and the level of IL-10 in lymph was significantly higher than that of blood(P〈0.05), however, the level of TGF-β in lymph were significantly lower than that of blood(P〈0.05) ; There was no significant difference in the percentage of CD4+ CD25+ Foxp3+ Treg in the blood and the levels of IL-10 and TGF-β in the plasma between control group and tolerance group(P〉0.05), indicating the imbalance both in quantity and function of the CD4+ CD25+ Foxp3+ Tregs in the lymph of bronchial asthmatic rat as well as the higher percentage of this type of cells in lymph than that in blood. In addition, the quantity of CD4+ CD25+ Foxp3+ Tregs in lymph and blood was increased. Accordingly, asthma could be avoided or its clinical symptoms relieved through the establishment of immunologic tolerance, and its establishment may be an effective way to treat asthma.
