摘要
从本实验室筛选的琼氏不动杆菌中获得褐藻胶裂解酶,酶解制备褐藻胶寡糖,用氧自由基吸收能力(ORAC)法测定抗氧化活性,并研究提高其抗氧化效果的方法。结果显示,该酶的最适酶解温度30℃,pH7.5。加入初始浓度为0.75%的海藻酸钠反应,2h达到最大降解率,且此时的平均聚合度(DP)为2;此时的抗氧化能力也达到最大值,平均ORAC值为1374.25μmol TE/g。反应体系中加入VC的平均ORAC值有所增加,且随浓度增加先增大后减少,在浓度为0.004g/L时达到最大值。实验表明:褐藻胶裂解酶制备的褐藻胶寡糖具有较强的抗氧化能力,且抗氧化效果与还原糖含量有关。VC保护反应能提高抗氧化能力,且与VC浓度有关。
Algino-oligosaccharide production from alginate was carried out by alginate lyase obtained from a newly isolated Acinetobacterjtmii. Oxy: gen radical absorbance capacity (ORAC) was used to study the capacity of antioxidation, and to find methods to improve the capacity. The results showed that the optimal temperature and pH of alginate lyase were obtained as 30℃ and pH 7.5, respectively. When alginate was added with an initial concentration of 0.75 %, the largest extent of the reaction happened at 2 h, and the average degrees of polymerization(DP) of alginate-degraded products was about 2 h. The antioxidant capacity also reached to maximum 1374.25 μmol TE/g. In Vc reaction systems, the antioxidant capacities were better than control, and the maximal level was 0.004 g/L. The results suggested that these products had the capacity of antioxidation and it had relation with DP. The antioxidant capacity could be improved by adding VC reaction.
