摘要
目的:总结国内外在乳腺癌中关于PTEN的研究进展,探索其对于乳腺癌治疗的指导作用。方法:利用PubMed数据库检索系统,以"乳腺癌、PTEN、AKT"为关键词,检索近5年的相关文献,共检索到英文文献104篇。纳入标准:1)PTEN来源和分子结构特征;2)PTEN发挥功能的几种途径,特别是PI3K/AKT途径;3)PTEN与乳腺癌治疗的关系。根据纳入标准,最后精选了43篇文章分析。结果:SHIP2、PIK3CA和BRCA1对PTEN的抑癌作用起负性作用,而BMP2、E-钙黏蛋白和p27kip1能促进PTEN的抑癌作用。PTEN表达决定乳腺癌细胞对于化疗药物doxo-rubicin、tamoxifen、ZD1839和trastuzumab的敏感性,却降低了对rapamycin的反应性。结论:虽然已经知道抑癌基因PTEN与乳腺癌密切相关,但其具体机制仍不清楚,需要进一步研究。
OBJECTIVE:To sum up the articles related to the tumor suppressor PTEN in the breast cancer,and explore its guiding role in breast cancer treatment.METHODS:The "breast cancer,PTEN,and AKT" were used as keywords,the relevant literature was retrived over the past five years,and a total of 104 English-language papers were retrieved by PubMed database retrieval system.The inclusion criteria: 1) PTEN sources and characteristics of its molecular structure;2) PTEN function in several pathways,especially in the PI3K/AKT way;3) The relationship between PTEN and breast cancer treatment.According to the inclusion criteria,finally 43 articles were analyzed.RESULTS:SHIP2,PIK3CA and BRCA1 play negative roles in the tumor suppressor role of PTEN,and BMP2,E-cadherin and p27^kip1 can promote the role of PTEN tumor suppressor.The different expressions of PTEN in some tissues are able to predict the sensitivity of breast cancer cells to chemotherapeutic drugs doxorubicin,tamoxifen,ZD1839 and trastuzumab,but reduce rapamycin responsiveness.CONCLUSION:There exists a close relationship between tumor suppressor PTEN and breast cancer,but the accurate menchanism is still unclear and need to be researched further.
