摘要 目的:探讨喉癌中PTEN、CyclinD1的表达及其与DNA含量的关系。方法:采用免疫组化S-P法和流式细胞术,检测PTEN/CyclinD1基因蛋白在60例喉鳞状细胞癌、15例喉乳头状瘤及10例正常黏膜的表达及DNA含量。结果:①PTEN蛋白表达定位于细胞质,PTEN蛋白阴性占18.3%(11/60),表达减低者占30%(18/60),阳性表达者占51.7%(31/60),PTEN与组织分化程度、临床分期相关。CyclinD1蛋白表达定位于细胞核,CyclinD1阴性占15%(9/60),表达减低者占30%(18/60),阳性占55%(33/60),CyclinD1与淋巴转移相关。PTEN表达与CyclinD1表达未显示有明显相关性。②喉癌的DI值高于对照组(P<0.05)。PTEN阳性的喉癌细胞DNA含量明显低于阴性组(P<0.05)。结论:PTEN、CyclinD1在喉癌的发生发展中起着不同程度的作用,它们的检测对喉癌的判定、预后和进一步治疗提供有效的依据。 Objective:To investigate the relationship between the expression of PTEN CyclinD1 and the DNA content in laryngeal carcinoma.Methods: expression of PTEN and CyclinD1 protein was measured with immunohistochemistry method and the DNA quantitative was measured with flow cytometry in 60 cases of laryngeal squamous cell carcinoma,15cases of larynx papilloma and 10 cases of normal laryngeal mucosa.Results:①PTEN,being show in the cytoplasm,was negative in 18.3 %,reduced in 30% and positive in 51.7%.PTEN expression level was correlated with differentiation degree and clinical stages.Immunoreacting to CyclinD1 was localized in the nuclei of cells,being negative in 15%、Reduced in 30% and being postiva in 55%.CyclinD1 expression level was correlated with lymphnode metastasis,There was no correlation between CyclinD1 expressionand PTENexpression.②The DNA index of laryngeal carcinoma group was higher than that of other groups(P〈0.05).The DNA index of laryngeal carcinoma whose expression of PTEN protein was positive was significantly lower than that of negative.While the DNA index was higher in negative expression of CyclinD1.Conclusion:This experiment suggests that PTEN and CyclinD1 may play certain roles in the oncogengsis and progression of laryngeal carcinoma.Detection of these gene pr ducts has positive offects on early diagnosis,malignancy determination prognosis and treatment of laryngeal carcinoma.Quamous
