摘要
目的研究肝脏超声剪切波速度与肝癌病灶内血管新生及存活素(Survivin)、第10号染色体同源丢失性张力蛋白基因(PTEN)的关系。方法选取行手术治疗的78例肝癌患者,取其术中肝癌组织及癌旁组织;测定癌组织与癌旁组织超声剪切波速度,根据检查结果进一步分为低剪切波速度35例、高剪切波速度43例。采用酶联免疫吸附法测定血管内皮细胞生长因子(VEGF)、基质金属蛋白酶9(MMP-9)水平,荧光定量PCR法检测Survivin、PTEN基因中mRNA表达量;分析剪切波速度与血管新生因子、Survivin、PTEN的相关性。结果肝癌病灶的剪切波速度高于癌旁病灶(P<0.05)。高剪切波速度病灶中VEGF、MMP-9水平高于低剪切波速度病灶(P<0.05)。高剪切波速度病灶中Survivin表达量高于低剪切波速度病灶,PTEN表达量低于低剪切波速度病灶(P<0.05)。经Spearman相关性分析,肝脏超声剪切波速度与病灶内血管新生因子VEGF、MMP-9水平及Survivin表达量呈正相关(P<0.05),与PTEN表达量呈负相关(P<0.05)。结论肝癌患者肝脏超声剪切波速度水平较高,且该水平与病灶内血管新生因子含量、Survivin和PTEN表达量密切相关。
Objective To investigate the relationship between liver shear wave velocity and angiogenesis,Survivin and phosphatase and tensin homolog deleted on chromosome ten(PTEN)in liver cancer tissues.Methods 78 patients with liver cancer underwent surgery were enrolled.The liver tissues and adjacent normal tissues were collected,and all patients received ultrasound examination.The results showed 35 cases of low shear wave velocity and 43 cases of high shear wave velocity.Enzyme-linked immunosorbent assay was used to detect the levels of vascular endothelial cell growth factor(VEGF)and matrix metalloproteinase-9(MMP-9),and fluorescent quantitative PCR method was used to detect the mRNA expression of Survivin and PTEN genes;Then the correlation of liver ultrasound shear wave velocity with angiogenesis,Survivin and PTEN was discussed.Results The shear wave velocity of liver cancer tissues was higher than that of adjacent normal tissues(P<0.05).The levels of VEGF and MMP-9 in high shear wave velocity lesions were higher than those in low shear wave velocity lesions(P<0.05).Survivin expression in high shear wave velocity lesions was higher than that in low shear wave velocity lesions,and PTEN expression was lower than that in low shear wave velocity lesions(P<0.05).Spearman correlation analysis showed that liver ultrasound shear wave velocity was positively correlated with the levels of angiogenesis factors VEGF,MMP-9 and Survivin expression in liver cancer tissues(P<0.05),and negatively correlated with PTEN expression(P<0.05).Conclusion Liver cancer patients have high shear wave velocity,which are closely related to the levels of angiogenesis factors and the expression of Survivin and PTEN.
