摘要
目的探讨全反式维甲酸(all-trans retinoic acid,ATRA)对体外培养的胃癌BGC-823细胞增殖和细胞凋亡的影响。方法不同浓度全反式维甲酸处理培养的胃癌BGC-823细胞后,采用MTT法检测细胞的增殖活性,采用TUNEL法检测细胞凋亡情况,采用RT-PCR检测细胞Survivin基因mRNA的变化。结果全反式维甲酸对胃癌BGC-823细胞的生长具有明显的抑制作用,其作用表现为剂量依赖性和时间依赖性(P<0.05)。TUNEL检测显示:以5μmol/L的全反式维甲酸对BGC-823细胞处理0、24、48、72h后细胞凋亡率逐渐增加(P<0.05),呈时间依赖性。RT-PCR检测显示BGC-823细胞Survivin基因的mRNA表达呈时间依赖性下调。结论全反式维甲酸对胃癌BGC-823细胞株具有明显的增殖抑制和诱导凋亡作用,其作用机制可能与下调Survivin基因表达有关。
Objective To observe the effect of ail-trans retinoic acid (ATRA)on proliferation and ap- optosis of human gastric cancer cell line BGC-823 and its mechanisms. Methods BGC-823 ceils were treated with different concentration of ATRA. The ceil proliferatiwas measured by methabenzthiazuron(MTI') assay and the apoptosis induced by ATRA was evaluated by TUNEL assay. The Survivin mRNA expression levels were de- tected by RT-PCR. Results ATRA inhibited growth of BGC-823 cells in a dose-dependent and time-dependent manner(P 〈0. 05). Cell apoptosis index increased gradually after exposure to ATRA for 24 h,48 h,72 h in a dose dependent manner( 1 ixmol/L) (P 〈 0.05 ). Survivin mRNA levels of BGC-823 cells treated with 1 p^mol/L of ATRA were reduced over time. Conclusion ATRA inhibits the proliferation and induces apoptosis in human gastric cancer cell line BGC-823. The effect mechanism is related to arrest the expression of Survivin gene.
