摘要
目的观察脓毒症大鼠的肺组织中c-FuP的变化情况,并探讨乌司他丁对c-FLIP水平的影响。方法将30只雄性Sprague-Dawley大鼠随机(随机数字法)分为假手术组(Sham组)、脓毒症模型组(CLP组)及乌司他丁治疗组(UTI组),每组10只。Sham组不结扎穿刺盲肠,余同CLP组;CLP组采用盲肠结扎并穿刺法造模;UTI组造模后立即经阴茎静脉注射乌司他丁注射液(50000U/kg)。12h后处死大鼠并采取肺组织,光镜下观察形态学变化;Western-Blot法检测c-FLIP蛋白表达,RT-PCR法检测c-FLIP基因表达。应用SPSS11.5进行统计学分析,多组比较采用方差分析,两两比较采用LSD-t检验。结果脓毒症组光镜下肺泡腔内中性粒细胞和大量红细胞渗出,c-FLIP蛋白和基因表达较假手术组显著降低(P〈0.05);用乌司他丁治疗后,光镜下病理改变减轻,c-FLIP蛋白和基因表达较脓毒症组显著升高(P〈0.05)。结论脓毒症中c-FLIP在肺组织中的表达下降,提示肺组织的细胞凋亡增加。乌司他丁能上调脓毒症大鼠肺组织中c-FLIP的表达。
Objective To explore the changes of c-FLIP in the lung of rats with sepsis,and to investigate the effect of Ulinastatin on the level of c-FLIP. Method Thirty male Sprague-Dawley rats were randomly ( random number) divided into 3 groups: sham operated group(Sham), sepsis group (CLP) and Ulinastatin treated group (UTI), n = 10,respectively. In Sham group, only laparotomy was performed. In CLP and UTI groups, cecal lig- ation and puncture was performed to induce sepsis. Ulinastatin (50 000 U/kg) was administered intravenously via penile vein in UTI group after operation. The same volume of nonnal saline instead of Ulinastatin was administered intravenously in Sham and CLP groups. The lungs of rats were harvested 12 hours after operation for determination of c-FLIP mRNA expressions assayed by using RT-PCR and cFLIP protein measured by Western blot. Statistical analysis was performed with SPSS version 11.5 software. ANOVA was used for multiple groups comparison, and t -test for paired comparison. Results Compared with Sham group, there was a significant decrease in c-FLIP pro- tein level and c-FLIP mRNA expression in lung ( P 〈 0.05). After treatment with Ulinastatin, the level of c-FLIP protein and mRNA expression obviously increased compared with CLP group ( P 〈 0.05). Conclusions The levels of c-FLIP decreased in lung of rats with sepsis, which suggests that pulmonary apoptosis might increase. Ulinastatin can increase the c-FLIP levels in the lung.
