摘要
目的:探讨内皮抑素基因治疗子宫内膜异位症裸鼠模型异位病灶中B细胞淋巴瘤/白血病-2(Bcl-2)蛋白和增殖细胞核抗原(PCNA)蛋白的表达。方法:将成模的子宫内膜异位症裸鼠模型60只随机分成3组:内皮抑素组(n=20)、空病毒组(n=20)、PBS组(n=20),应用免疫组化法检测裸鼠模型异位病灶中Bcl-2和PCNA的表达。结果:3组中的腺体均萎缩,但是内皮抑素组的腺体萎缩及血管减少较空病毒组和PBS组明显。给予治疗后,Bcl-2和PCNA均在腺体中表达,Bcl-2在内皮抑素组的阳性表达率明显低于在空病毒组的阳性表达率及在PBS组的阳性表达率,有显著性差异(P<0.05),Bcl-2在空病毒组与PBS组的阳性表达率无差异(P>0.05);PCNA在内皮抑素组的阳性表达率明显低于在空病毒组的阳性表达率及在PBS组的阳性表达率,有显著性差异(P<0.05),PCNA在空病毒组与PBS组的阳性表达率无差异(P>0.05)。结论:内皮抑素能够治疗子宫内膜异位症。
Objective： To study the expression of Bcl-2 and proliferating cell nuclear antigen（PCNA） in ectopic endometrum of nude mice. Methods： Sixty nude mice with endometriosis were divided randomly into three groups：the endostatin group （n=20）,the group without endostatin （n=20）,the PBS group （n=20）. Using SP immunohistoehemical technique, Bcl-2 and PCNA were assessed in ectopic endometrium of model mice. Results： The gland in three groups withered, the blood vessel in three group reduced;and the endostatin group gland reduced obviously.Bel-2 and PCNA were both located in the gland through curing. The positive rate of expression about Bel-2 protein in the endostatin group was significantly lower than that in the group without endostatin（P〈0.05） and in the PBS group （P〈0.05）.The positive rate of expression about Bcl-2 protein in the group without endostatin was not significantly different from that of the PBS group （P〉0.05）. The positive rate of expression about PCNA protein in the endostatin group was significantly lower than that in the group without endostatin （/~〈0.05） and in the PBS group （P〈0.05）.The positive rate of expression about PCNA protein in the group without endostatin was not significantly different from that of the PBS group （P〉0.05）. Conclusion： Endostatin can cure endometriosis.
