摘要
目的观察益气解毒方原方及其拆方对人鼻咽癌细胞CNE2 nm23-H1表达活性及运动潜能的影响,分析nm23-H1活性与瘤细胞迁徙潜能的关系,从细胞迁徙干预机制探讨其对鼻咽癌细胞转移潜能的抑制效应。方法制备益气解毒方原方及其拆方的大鼠药物血浆,MTT法确定药物血浆的IC50浓度,分别作用于体外培养的CNE2细胞;划痕试验检测细胞迁徙潜能,免疫细胞化学法检测nm23-H1基因表达活性,比较分析各项检测指标的组间差异。结果与正常大鼠血浆组比较,益气解毒方原方及其拆方药物血浆分别作用24h后,细胞迁徙数和迁移距离均显著降低(P<0.05);各组药物血浆分别作用12h、24h后,CNE2细胞nm23-H1表达水平显著升高(P<0.05),尤以原方和解毒组分作用明显,而益气组分和养阴组分则否。结论益气解毒方促进抑癌基因nm23-H1的表达活性并进而影响瘤细胞迁徙运动能力的生物力学机制,可能是该方抑制鼻咽癌细胞转移潜能的药理效应机制之一。
Objective To investigate the inhibiting effects of compound Chinese herbal medicine Qi-Boosting Toxin-Resolving Formula (QBTRF) and its composing components on the metastatic potentiality of nasopharyngeal carcinoma (NPC) cells via analyzing the action of these herbals on the expressive activity of nm23-Hl and cell migration in a consideration on the association of nm23-H1 expression level with the moving potentiality of NPC cells in the view of the mechanism of cellular moving intervening. Methods Medicine-containing plasma was prepared with the original formula of QBTRF and its composing components in rats respectively at first. Then, IC50 concentration of medicine-containing plasma was determined in vitro with the procedures of MTT, and each kind of plasma was added into the culturing system of CNE2 cells respectively. Following this step of experiment, the ability of cell moving was determined in a way of streak culturing assay, and the expressive activity of nm23-H1 was detected by immunohistochemical assay. Results As compared with that of healthy controlling rats, the numbers and reached distances of migrating cells were significantly reduced (P~ 0.05) when CNE2 cells cultured for 24 hours with the plasmas of original formula and its composing components respectively. Meanwhile, the expressive levels of nm23-H1 in these cells were obviously increased when cultured for 12 and 24 hours with the plasmas of some of these components respectively (P〈0.05), especially when that of the original formula and the composing component of toxin-resolving was used but not when that of Qi-boosting and Yin-nourishing components cultured with. Conclusions QBTRF holds definitely promoting effect on the expressive activity of nm23-H1 and gives rise inhibiting effect on the moving ability of CNE2 cells in turn via the network mechanism of biological dynamics. This may be one of the pharmacological affecting mechanisms for QBTRF to inhibit the metastatic potentiality of NPC cells.
