摘要
目的研究姜黄素对水浸束缚应激(water—immersion restraint stress,WRS)大鼠胃壁细胞形态改变的影响。方法60只健康SD大鼠随机分为姜黄素实验组、WRS对照组和空白对照组,每组20只。分别给予0.5%姜黄素溶液、生理盐水灌胃(后2组),2次/d,连续7d,用WRS制备大鼠应激性溃疡(stress ulcer,SU)模型。处死大鼠,应用免疫荧光方法标记壁细胞H^+/K^+-ATP酶后,利用荧光显微镜及激光共聚焦扫描显微镜观察3组大鼠胃壁细胞形态变化的差别。结果姜黄素实验组大鼠SU模型胃黏膜出血灶数、溃疡形成数与对照组比较差异有统计学意义(P〈0.05),胃分泌期胃壁细胞(网格样细胞)数与对照组比较差异有统计学意义(P〈0.05)。结论姜黄素能降低WRS大鼠胃黏膜分泌型胃壁细胞的形成,从而预防应激性溃疡的发生。
Objective To investigate effect of curcumin on morphology of gastric parietal cells in rats with water-immersion restraint stress (WRS). Methods Sixty SD rats were randomly divided into 3 groups: the eurcumin experimental group, the control group and the blank group. In the experimental group, rats were gavaged with 0.5% cureumin solution twice a day for a duration of 7 days. The other 2 groups were gavaged with physiologic saline solution. The stress ulcer (SU) rat model was developed with the WRS method. After the rats were sacrificed, H ~/K + -ATPase of gastric parietal cells was labeled with fluoroimmunoassay, and changes in morphology of gastric parietal cells were observed, categorized and counted with fluorescence microscope and confocal laser scanning microscope for the 3 groups. Results The number of hemorrhagic foei in gastric mucosa and the number of ulcer for the curcumin experimental group were all significantly lower than those of the control group ( P 〈 0. 05 ), and the number of secreting parietal ceils for the experimental group was also significantly lower than that of the control group ( P 〈 0.05 ). Conclusions Curcumin could decrease the number of secreting parietal cells and prevent the occurrence of gastric stress ulcer.
