摘要 目的:研究翠云草总黄酮对结肠癌HT-29细胞环氧化酶-2(COX-2)mRNA表达的影响。方法:以翠云草总黄酮作用于人结肠癌HT-29细胞,用倒置显微镜观察细胞形态学的变化;并设总黄酮高中低剂量组和阴性对照组共4个组别,采用逆转录聚合酶链反应(RT-PCR)法检测给药后癌细胞COX-2 mRNA表达的水平,用电泳凝胶成像自动分析系统检测各扩增带的产物含量。结果:与阴性对照组比较,翠云草总黄酮可以在mRNA水平上抑制COX-2的表达,且呈量效关系。结论:翠云草总黄酮能在mRNA水平上抑制COX-2,进而抑制COX-2蛋白质表达,这可能是翠云草抗肿瘤作用的机制之一。 bjective : To investigate the action of total flavones from Selaginella uncinata ( Desv. ) Spring on cyclooxygenase-2 (COX-2) mRNA expression in carcinoma of colon cell line HT-29 ,and explore related molecular mechanism thereby. Method: Total flavones from Selaginella uncinata was used on TH-29 cells and there were four groups including high-dose group, midst-dose group, low-dose group and negative control group. The morphological change of the cells was observed by inverted microscope. The expressions of COX-2 mRNA were measured using reverse transcription polymerase chain reaction (RT-PCR) method. Caraphoresis gel photo auto- analysis system was used to detect the products. Result: Comparing with the negative control group, the total flavonoids groups could significantly inhibit the expressions of the COX-2 mRNA level in a dose-dependent manner. Conclusion: It can be concluded that the mechanisms may be that the total flavonoids can inhibit COX-2 in mRNA level. Then it can inhibit COX-2 protein expression. It may be one of the anti-tumor mechanisms of Selaginella uncinata.
