摘要
目的建立测定复方葛酮颗粒中葛根素含量的高效液相色谱(HPLC)法。方法以WatersC18柱(250mm×4.6mm,5μm)为色谱柱,流速1.0mL/min,检测波长250nm,甲醇-水-醋酸(25∶75∶1)为流动相。结果葛根素进样量的线性范围为0.048~1.92μg(r=0.9999,n=6),平均回收率为99.12%,RSD为1.62%(n=6)。结论HPLC法简便、准确,重现性好,可用于复方葛酮颗粒的质量控制。
Objective To determine the content of puerarin in Compound Getong Granules by HPLC. Methods The HPLC method was used with the Waters C18 column (250 mm ×4.6 mm, 5 μm). The mobile phase was methanol-water- glacial acetic acid (25:75:1). The detection wavelength was at 250 nm and the flow rate was 1.0 mL/min. Results Puerarin had the good linear relationship in the concentrations between 0. 048 - 1.92μg(r=0. 999 9, n =6) with the average recovery rate of 99. 12% (RSD = 1.62% ). Conclusion The method is simple, accurate, reliable and high sensitive, which can be regarded as the quality control method of Compound Getong Granules.
