摘要
目的:观察CpG寡脱氧核苷酸(ODN)对1型糖尿病患者与健康志愿者外周血单个核细胞(PBMC)γ干扰素(IFN-γ)、白细胞介素(IL)-12以及IL-10表达的影响。方法:将1型糖尿病患者与健康志愿者的PBMC根据刺激物不同分为对照组、CpG组(CpGODN 2 nmol·mL-1)。用逆转录-聚合酶链反应(RT-PCR)法检测PBM CIFN-γmRNA、IL-12mRNA和IL-10mRNA的表达。结果:1型糖尿病患者IFN-γmRNA和IL-12mRNA的表达明显低于健康志愿者;1型糖尿病患者与健康志愿者PBMC经CpGODN刺激后,IFN-γ mRNA和IL-12mRNA的表达均高于对照组(P<0.01,P<0.01),IL-10mRNA的表达与对照组相似(P>0.05)。结论:1型糖尿病患者PBMC表达IFN-γ和IL-12下降,CpGODN可促进其表达,增强其免疫调节能力。
OBJECTIVE: To evaluate the effects ofCpG oligonucleotide (ODN) on the expression of interferon γ (IFN-γ), interleukin (IL)-12 and IL-10 of peripheral blood mononuclear cells (PBMC) in patients with type 1 diabetes meUitus (TIDM) or healthy volunteers. METHODS: PBMC were isolated from T1DM patients or healthy volunteers were assigned to either control group or GpG ODN group (2 nmol·mL^-1) based on the difference in stimulants. The expression of IFN-γ mRNA, IL-10 mRNA and IL-12 mRNA in PBMC were detected by RT-PCR. RESULTS: The expression of IFN-γ mRNA and IL-12 mRNA in T1DM patients were significantly lower than that in health volunteers. In CpG ODN-treated patients (TIDM patients and healthy volunteers) had higher expression levels of IFN-7 mRNA and IL-12 rnRNA than that in control group (P〈0.01, P〈0.01), but the expression level of IL-10 mRNA was similar to that in control group (P〉0.05). CONCLUSION: Lowered expression of IFN-γ and IL-12 in PBMC of T1DM patients was noted, but CpG ODN treatment can promote the expression of IFN-γ and IL-12 and enhance the immunoregulation ability of T1DM patients.
