摘要
目的探讨Caspase-12 mRNA在淋巴滞留性脑病大鼠模型脑皮层及海马中的表达情况,及其与神经元凋亡之间的关系。方法健康成年雄性Wistar大鼠,随机分为手术组、假手术组和对照组,手术组建立淋巴滞留性脑病实验动物模型,在脑淋巴阻滞后不同时间点(1、3、5、7、14d)取颞顶皮层、海马区脑组织,用半定量RT-PCR方法观察淋巴滞留性脑病大鼠损伤后Caspase-12 mRNA的表达情况。用假手术组、对照组观察并对比上述指标。结果在颞顶皮层,Caspase-12 mRNA的表达在术后1d即升高,3d时达到高峰,5d时开始下降,但仍高于正常对照组及假手术组,且有统计学意义(P<0.05),术后7d、14d降至接近正常对照及假手术组水平。在海马,淋巴阻滞后3d内Caspase-12 mRNA的表达升高,且以第一天时为最高,5d后逐渐接近正常对照组及假手术组。结论凋亡的发生可能与内质网应激有关。
Objective To investigate the expression of Caspase - 12 mRNA in the rat model of lymphostatic encephalopathy and their relationships between neuronal apoptosis and lymphostatic encephalopathy. Methods 88 male adult Wistar rats were randomly divided into 3 groups: operation group, sham-operation group and control. We used operation group to establish the rat model of lymphostatic encephalopathy, after a series of time points ( 1 d, 3 d, 5 d, 7 d, 14 d), animals were sacrificed, temporoparietal cortex and hippocampal tissue were isolated and stored in - 80℃ for later use. Semi-quantitive RT-PCR technique was used to explore the expression of Caspase - 12 mRNA. Results In temporopariental cortex, Caspase - 12 mRNA expression rose at 1 day and reached the highest level at 3 days after operation. Then it began to decline. But it showed a significant difference compared with sham-operation group and control. In hippocampus, we could see a higher expression of Caspase - 12 mRNA from 1 to 3 days after operation. The highest occurred at the first day after operation. Conclusion In the rat model lymphostatic encephalopathy, neuronal apoptosis is probably related to the activation of endoplasmic reticulum pathway.
