半胱氨酸天冬氨酸蛋白酶3 mRNA在淋巴滞留性脑病大鼠脑皮质及海马的表达 被引量:2

摘要 目的:探讨半胱氨酸天冬氨酸蛋白酶3mRNA在淋巴滞留性脑病大鼠脑皮质及海马中的表达情况及其与神经元凋亡之间的关系。方法:选取成年雄性Wistar大鼠88只,随机分为脑淋巴引流阻断组40只、假手术组40只及正常对照组8只。脑淋巴引流阻断组和假手术组分为术后1,3,5,7,14d5个时间点,每个时间点8只。脑淋巴引流阻断组建立淋巴滞留性脑病动物模型,在损伤后各时间点取颞顶皮质、海马区脑组织。假手术组只分离和暴露淋巴结,但不结扎淋巴管,亦不摘除淋巴结。用半定量反转录-聚合酶链反应方法观察淋巴滞留性脑病大鼠损伤后半胱氨酸天冬氨酸蛋白酶3mRNA的表达情况。结果:88只大鼠均进入结果分析。①在颞顶皮质,Caspase3mRNA在淋巴阻塞后3d内表达上调,以第1天表达最高,淋巴阻塞后3d内Caspase3mRNA表达比假手术组及对照组高,且有统计学意义(F=29.892,P=0.001<0.01)。淋巴阻塞3d后表达接近正常对照及假手术组。对照组与假手术组之间差异无统计学意义。②在海马,Caspase3mRNA较颞顶皮质表达上调更明显,以第1天表达最高,淋巴阻塞后5d内Caspase-3mRNA表达比假手术组及对照组高,差异有统计学意义(P<0.05),5d后表达接近正常对照及假手术组。结论:在淋巴滞留性脑病大鼠模型中神经元凋亡的发生可能与半胱氨酸天冬氨酸蛋白酶-3mRNA的激活有关。 AIM: To investigate expression of easpase 3 mRNA in the cerebral cortex and hippocampus of rat models with lymphostatic encephalopathy and the relationship between neuronal apoptosis and lymphostatic encephalopathy. METHODS: A total of 88 male adult Wistar rats were selected and randomly divided into 3 groups: operation group (n=40), sham operation group (n=40) and normal control group (n=8). The rats in the operation group and sham operation group were assigned into 1, 3, 5, 7 and 14 days time points after operation with 8 rats in each group. The rat models with lymphostatic encephalopathy in the operation group were established. Temporoparietal cortex and hippocampal tissues were isolated at each time points after injury. Only glandula lymphatica in the sham operation group was removed and exposed, but the lymphatic vessel was not ligated, and the glandula lymphatica was not extracted. The expression of caspase-3 mRNA was observed after injury in rats with lymphostatic encephalopathy with semiquantitative reverse transcription-polymerase chain reaction. RESULTS: Totally 88 rats were involved in the result analysis. ①Caspase-3 mRNA was up-regulated in temporoparietal cortex within 3 days after lympha block. The expression was the highest in the 1" day. The expression of caspase-3 mRNA was higher than that in the sham operation group and control group within 3 days, which had statistical significance (F=29.892,P=0.001 〈 0.01 ). The expression after 3 days block was near to that in the normal control group and sham operation group. There was no statistical significance between control group and sham operation group. ②In the hippocampus, the expression of caspase-3 mRNA was more significantly up-regulated than that in temporoparietal cortex. The expression was the highest in the 1" day.
