摘要
目的观察三七总皂甙对单侧输尿管梗阻(UUO)大鼠肾间质纤维化的影响。方法将雄性SD大鼠80只随机分为4组:UUO组(UUU,n=20)、假手术组(SOR,n=20)、三七总皂甙组(PNS,50 mg/(kg.d),n=20)和氯沙坦组(ARB,20mg/(kg.d),n=20)。从术后24小时开始,治疗组分别予PNS及ARB治疗,连续14天。于术后3、7、14天分别随机处死各组大鼠。行Masson染色观察肾间质病理学改变;化学比色法测定肾组织匀浆中羟脯氨酸的含量;采用Western蛋白印迹从蛋白水平观察金属蛋白酶1组织抑制剂(Tissue inhibitor of metalloproteinase,TIMP-1)的表达情况。荧光定量PCR(Real-time PCR)从mRNA水平观察胶原Ⅰ(Collagen I,Col-Ⅰ)和纤维粘连蛋白(Fibronectin,FN)的表达。结果UUO组肾脏呈进行性肾间质纤维化及肾间质胶原沉积的改变(P<0.05)。PNS组与UUU组比较均不同程度地减轻了胶原在肾间质的沉积,改善了肾脏病理损害,抑制了TIMP-1的表达(P<0.05)。结论PNS对UUO所致的大鼠肾脏纤维化的形成有明显的抑制作用,其机制可能与抑制TIMP-1的表达有关。
[Objective] To study the effect of panax notoginseng saponins on renal interstitial fibrosis caused by unilateral ureteral obstruction (UUO) in rats. Methods 80 healthy Sprague-Dawley rats were randomly divided into 4 groups, ineluding UUO group (UUO, n=20), sham-operation group (SOR, n=20), UUO with Panax notoginseng saponins group (PNS, 50mg/(kg · d), n= 20) and UUO with Losartan group (ARB, 20mg/(kg · d), n = 20). The treatment groups were given either PNS or losartan for 14 days. The rats were saerificed at day 3, 7 and 14 after the surgery, respectively. The histological changes of renal interstitial tissues were observed by Masson staining. Total kidney collagen content was determined by measuring the amount of hydroxyproline. The mRNA of collagen I and fibroneetin were reverse transcribed and quantified by real-time PCR. The Protein of tissue inhibitor of metalloproteinase(TIMP-1) expression was assessed by western blot analysis. Results UUO was found to induce fibrosis and the accumlation of collagen in the obstructed kidney ( P〈0. 05). Administration of PNS improved tubulointerstitial damage and collagen matrix accumulation in dif- ferent degree. The expression of the TIMP-1 at protein levels were significantly increased in the UUO group (P〈0. 05), which was also suppressed by treatment with PNS. Conclusion PNS could evidently inhibit renal interstitial fibrosis via suppressing the expression of TIMP-1 in rats with UUO.
