摘要
目的探讨十味芪黄益肾方对腺嘌呤所致肾纤维化大鼠模型TGF-β1/Smads/BMP-7信号通路的影响。方法选取40只6周龄SPF级大鼠,体重(200±30)g,采用腺嘌呤灌胃的方法制备肾纤维化大鼠模型,并分为芪黄益肾方组,模型组、正常对照组和TGF-B抑制剂组。通过灌胃的方法给大鼠服用十味芪黄益肾方4周,每隔一周检测大鼠体重、血清肌酐(Scr)、尿素氮(BUN)和血红蛋白(Hb)的含量。使用Westen Bolt和RT-PCR定性、定量检测大鼠TGF-β1、Smad2、Smad 6及BMP-7、BMP-7RII、PDGFR-α、PDGFR-β、PDGF-A和PDGF-B蛋白表达情况,将大鼠肾脏组织染色后,观察大鼠肾脏组织的病理改变及肾间质纤维化情况。结果与模型组大鼠比较,十味芪黄益肾方组和TGF-B抑制剂组大鼠体重显著增加,Scr、BUN显著降低,Hb显著升高;TGF-β1、Smad 2蛋白表达量显著降低,Smad 6、BMP-7和BMP-7RII蛋白表达量显著升高(均P<0.05)。结论十味芪黄益肾方通过下调TGF-β1、Smad 2蛋白表达量,上调BMP-7和BMP-7RII蛋白表达量,从而抑制了TGF-β1信号向细胞核内转导的通路,缓解了腺嘌呤所致肾纤维化的进程。
ObjectiveTo explore the effects of Shiwei Qihuang Yishen recipe on TGF-β1/Smads/BMP-7 signaling pathway in rat models with renal fibrosis induced by adenine. MethodsRat models of renal fibrosis were prepared by adenine gavage, and were divided into Qihuang Yishen recipe group, model group, normal control group and TGF-B inhibitor group. Rats were given Shiwei Qihuang Yishen recipe for 4 w by intragastric administration. The body weight and contents of serum creatinine (Scr), urea nitrogen (BUN) and hemoglobin (Hb) were measured every other week. The expressions of TGF-β1, Smad 2, Smad 6, BMP-7 and BMP-7 RII proteins were detected by Western Bolt and RT-PCR qualitatively and quantitatively. The kidney tissues of rats were stained and the pathological changes and renal interstitial fibrosis of rat kidney tissues were observed and recorded. ResultsCompared with model group, the body weight in Shiwei Qihuang Yishen recipe group and TGF-B inhibitor group was significantly increased, and the serum creatinine (Scr) was significantly decreased. The serum urea nitrogen (BUN) was significantly decreased. The hemoglobin (Hb) was significantly increased. The TGF-β1 protein expression was significantly decreased. The Smad 2 protein expression was significantly decreased. The Smad 6 protein expression was significantly increased. The expression levels of BMP-7 and BMP-7 RII proteins were significantly increased. ConclusionShiwei Qihuang Yishen recipe can inhibit the transduction of TGF-β1 signal into the nucleus by down-regulating the expression of TGF-β1 and Smad 2 proteins and up-regulating the expression of BMP-7 and BMP-7 RII proteins, thus relieving the progression of renal fibrosis caused by adenine.
