表面活性蛋白-A和表面活性蛋白-D在人正常肾组织的表达与分布

Expressions and Distribution of Surfactant Protein-A and-D in Normal Human Kidney

摘要 目的:研究表面活性蛋白-A(SP-A)和表面活性蛋白-D(SP-D)在人正常肾组织的表达及表达部位。方法:收集5例正常的人肾组织,培养人肾小管上皮细胞(HK-2),同时收集5例正常的人肺组织作为对照。应用免疫组化技术检测SP-A和SP-D蛋白在人正常肾组织的表达部位;应用RT-PCR检测SP-A和SP-D mRNA在正常肾组织和HK-2细胞中的表达。结果:免疫组化方法检测到SP-A和SP-D主要表达在皮质部的近曲和远曲小管;RT-PCR结果显示,在人的正常肾组织和HK-2细胞中存在SP-A和SP-D mRNA表达。结论:与肺组织有相同内胚层来源的肾组织可表达SP-A和SP-D。 Objective: To investigate the expressions and distribution of surfactant protein-A(SP-A) and SP-D in normal human kidney. Methods: The expressions of SP-A and SP-D genes in human renal tubule epithelial cells (HK-2) in vitro and in five normal human kidneys were detected by RTPCR, and the distribution of SP-A and SP-D proteins were assayed by immunohistochemistry in five normal human kidney tissues, and compared with those in normal human lung tissues. Results: Immunohistochemical staining showed the distributions of SP-A and SP-D proteins were in distal and proximal convoluted tubule of human normal renal tissue; SP-A and SP-D mRNA could be detected by RT-PCR in both normal human kidney and HK-2 cells. Conclusion: SP-A and SP-D proteins and mRNA may be expressed in normal human kidney.
