摘要
目的探讨不同浓度阿奇霉素对体外流感嗜血菌成熟生物膜的破坏作用。方法通过结晶紫染色、扫描电镜及菌落计数观察体外不同浓度阿奇霉素对成熟生物膜的黏附性、基本结构及膜内活菌数的破坏作用。结果结晶紫染色结果显示64~1/64MIC浓度阿奇霉素作用两株流感嗜血菌生物膜吸光度值均比阳性无药对照组低,差异有统计学意义,高于1MIC浓度比亚MIC浓度吸光度值低;10MIC阿奇霉素作用流感嗜血菌后可见生物膜结构松散,细菌清除后呈网格状改变;菌落计数结果显示,随着阿奇霉素的浓度逐渐增高,流感嗜血菌生物膜内的活菌数明显减少,且在1/32 MIC时对膜内细菌仍有抑制作用,同时当阿奇霉索浓度高达8MIC时仍不能完全清除膜内细菌。结论阿奇霉素可影响生物膜黏附性、破坏生物膜结构,并可减少生物膜内活菌数。
OBJECTIVE To determine the activity of azithromycin on bacterial biofilms formed by Haemophilus Influenzae in vitro. METHODS The destruction of different concentrations azithromycin on H. influenzae biofilms was evaluated through crystal violet assay and scanning electron microscopy, viable cells counting in biofilms was also carried out. RESULTS Optical density of biofilms and viable cells counting was significantly decreased at the concentration of 64 - 1/64 MIC for azithromycin, and biofilm structure was destroyed by azithromycin at concentration of 10 folds of MIC. CONCLUSIONS Azithromycin is able to interfere with adhesion, damage the structure of biofilm, and reduce viable ceils counting at high concentrations.
