中药红藤合剂灌肠对葡聚糖硫酸钠诱导的小鼠溃疡性结肠炎的影响

Effect of Coloclysis with Chinese Crude Drug on the Colitis Mousse Induced by Deztran Sulphate Sodium

摘要 目的探讨中药红藤合剂灌肠对葡聚糖硫酸钠(DSS)诱导小鼠结肠炎的影响,为中药灌肠的临床应用提供实验依据。方法给昆明小鼠连续饮用5%的葡聚糖硫酸钠13天,造成小鼠结肠炎模型。实验分为对照组(未处理)、模型组(DSS处理)和实验组(造模后红藤合剂灌肠)。病理切片检测小鼠局部肠道病理变化;ELISA法检测局部肠道组织细胞因子分泌情况;WesternBlot法检测肠道组织IκBα、NF-κB表达。结果模型组小鼠疾病活动指数、病理评分均明显高于对照组,局部组织细胞因子IL-12p、TNF-α明显上升,抑炎因子IL-10表达下降,IκBα表达减弱,NF-κB表达上调,且中药治疗组可以有效地缓解临床症状,调节局部肠道组织IκBα、NF-κB细胞因子的表达。结论中药红藤合剂灌肠对DSS诱导的结肠炎小鼠有较好的治疗作用,而这种作用可能是通过对肠道IκBα、NF-κB的表达产生影响,并进而调节局部细胞因子的表达来达成的。 Objective To explore the effect of coloclysis with Chinese crude drug (CCD) on the colitis mouse induced by dextran sulphate sodium(DDS) and provide the experiment evidence for the clinical use. Methods The colitis mouse model were induced by adding 5% DDS into the drinking water after 13 days. After the coloclysis with CCD for 7 days, the mouse was killed and pathological sections were used to detect the changes of local intestinal mucosa,ELISA was used to detect the cytokine secreted of intestinal mucosa and Western Blot was used to detect the protein of IκBα,NF-κB. Results The DAI scores and histological scores of the model group were significantly higher than the control group,the level of IL-12 and TNF-α,the expression of NF-κB in the local intestinal mucosa of model group increased ,the level of IL-10 and the expression of IκBα decreased( all P 〈 0. 01 ). The coloclysis with CCD can relief the clinical symptoms,adjust the cytokine secretion and the expression of IκBα, NF-κB. Conclusion Coloclysis with CCD is much helpful to the colitis mouse induced by DDS. This therapeutic effect maybe act though the expression of IκBα, NF-κB and the adjustment of the cytokiue secretion.
