骨桥蛋白在胰腺癌组织芯片中的表达及其临床意义 被引量:1

Expression and Clinical Significance of Osteopontin in Pancreatic Carcinoma Using Tissue Microarray

摘要 目的探讨胰腺癌组织中骨桥蛋白(OPN)的表达及其与胰腺癌临床病理特征和预后的相关性及意义。方法将88例胰腺癌组织和11例正常胰腺组织制成组织芯片,采用二步法免疫组化方法检测OPN在胰腺癌中的表达,并对随访2个月~6年的53例作了生存分析。结果胰腺癌中OPN阳性表达54例(61.36%)。OPN表达和胰腺癌的临床TNM分期晚、淋巴结转移存在显著相关性(P<0.01),与肿瘤病理分级、部位和大小无关(P>0.05)。对随访的53例进行单因素、多因素COX分析显示,OPN表达与预后有关(P<0.05)。结论OPN在胰腺癌中过表达可能参与了胰腺癌的发生、发展和转移。OPN是胰腺癌有意义的预后指标。 Objective To investigate the expression of phosphorylated osteopontin (OPN) in pancreatic ductal adenocacinoma(PDAC) by high-throughput tissue microarray technique,as well as its relationship with the clinicopathologic features besides the prognostic significance of OPN expression in PDAC. Methods The pancreatic cancer tissue microarray was composed of 88 specimens of the pan-creatic adenocarcinoma and 11 normal pancreatic tissues as the control subjects. Two steps immunohis-tochemistry method was performed to examine expressions of OPN. In addition,53 of the 88 cases were followed-up from 2 months to 6 years,in which the survival was analyzed. Results Positive expression of OPN in tumors was showed 54(61.36%). The expression of OPN was significantly associated with late TNM stage and lymph node metastasis(P〈0.01),but there was no distinct difference found in tumor location,size and pathologic grade (P〉0.05). Univariate and multivariate analysis revealed that OPN is a prognostic indicator for PDAC. Conclusion The pathogenesis,development and metastasis of pancreatic cancer is associated with OPN. The expression of OPN is a significant prognostic indicator for PDAC.
