摘要
目的研究雷公藤甲素(TP)是否具有抑制血管平滑肌细胞(VSMCs)增殖并诱导其发生凋亡的作用,为临床开发新型药物洗脱支架提供实验依据。方法主动脉SMCs体外培养和流式细胞仪检测,DNA凝胶电泳。结果TP组与阳性对照地塞米松相比,凋亡率有显著提高;TP干预组凝胶电泳谱型呈凋亡特征性梯状条带。结论TP可抑制指数生长期SMCs增殖并诱导其凋亡。
Objective To access the effects of triptolide on the proliferation and apoptosis of vascular smooth muscle cells(VSMCs), and to screen a new drug for the drug-eluting stents. Methods Aortal SMCs were cultured in vitro and their cell cycles were detected by flow cytometry. DNA gel electrophoresis was carried out to measure its spectral pattern. Results The apoptosis rate in triptolide group was significantly higher than that in the positive group. DNA ladders were also found in the treated groups. Conclusion This study demonstrates that triptolide can inhibit the proliferation and induce apoptosis in VSMCs.
