摘要
目的研究雷公藤甲素对雄性性成熟期大鼠附睾功能及精子运动的影响,探讨雷公藤甲素生殖毒性的作用机制。方法将2月龄健康清洁级Sprague-Dawley雄性大鼠40只随机分为对照组(0.5%的羧甲基纤维素钠)及低、中、高雷公藤甲素染毒组(染毒剂量分别为25、50、100μg/kg),经口灌胃染毒,每天1次。连续染毒50d后,取双侧附睾称重,并计算附睾系数。唾液酸含量的测定采用5-甲基苯二酚法,蛋白含量的测定采用考马斯亮兰法,肉毒碱含量的测定采用酶联免疫吸附法。采用计算机辅助精子分析(CASA)系统评价左附睾尾精子动力学参数。结果各染毒组大鼠体重间比较,差异无统计学意义(P>0.05)。与对照组相比,中、高剂量组唾液酸含量和高剂量组附睾脏器系数和肉毒碱含量均较低,差异有统计学意义(P<0.05或P<0.01)。与对照组比较,高剂量组精子曲线速度(VCL)、平均路径速度(VAP)、侧摆幅度(ALH)、鞭打频率(BCF)和中、高剂量组精子直线速度(VSL)、直线性(LIN)、前向性(STR)均较低,差异有统计学意义(P<0.05或P<0.01)。结论雷公藤甲素可以引起附睾功能损伤,继而影响附睾尾精子运动速度与线性方向。
Objective To investigate the effect of triptolide on epididymal function and sperm motility parameters in adult male rats,to explore the potential mechanism of triptolide in reproductive toxicity. Methods Forty male SD rats of clean grade aged 2 months were randomly divided into the control group and triptolide groups, the rats were treated with triptohde at the doses of 25,50 and 100 μg/kg respectively by gavage ,once a day,for 50 consecutive days. All rats were sacrificed on the 50th day and the epididymal coefficient was determined. The fight epididymis were collected to determine the contents of sialic acid by spectrophotometry ,protein by Bradford and carnitine by ELISA,while the left epididymis were collected to evaluate eauda epididymal sperm motility. Results The body weight of exposed rats were not obviously changed (P〉0.05).Compared with control group, contents of sialic acid in the high and moderate dose groups, epididymal coefficient and contents of carnitine in the high dose group reduced significantly(P〈0.05 or P〈0.01 ) and the sperm motility parameters such as curvilinear velocity(VCL), average path velocity(VAP), amplitude of lateral head displacement( ALH) and beat cross frequency(BCF) in the high dose group, straight-line velocity (VSL), percentage linearity (LIN) and percentage straightness (STIR) in the high and moderate dose groups reduced too (P〈0.05 or P〈0.01 ). Conclusion Triptolide may cause damage in the epididymis,which can present adverse effects on cauda epididymal sperm motility.
