摘要
以对羟基苯基丙烯酸为原料,通过羟甲基化得到3-羟甲基-4-羟基苯基丙烯酸,然后对3-羟甲基-4羟基-苯基丙烯酸的双键加成上羟基,再把羧酸还原成醛基,然后与4-苯氧丁基己胺缩合得到亚胺,加氢还原得到目标产物。
The Salmeterol was synthesized through 3-(4-hydroxyphenyl)acrylic acid as started materials. 3-(4-hydroxy-3-(hydroxymethyl)phenyl)acrylic acid was prepared from 3-(4-hydroxyphenyl)acrylic acid through methyloated reaction. Then 3-(4-hydroxy-3-(hydroxymethyl)phenyl)acrylic acid was got from H2O addition reaction wtih double bond to get derivative 3-hydroxy-3-(4-hydroxy-3-(hydroxymethyl)phenyl)propanal. At last, the 3-hydroxy-3-(4-hydroxy-3- (hydroxymethyl)phenyl)propanal react with amino and addition with H2 to get the target compound.
