摘要
目的观察红景天胶囊对大鼠局灶性脑缺血再灌注梗死区胶质纤维酸性蛋白(GFAP)表达的影响。方法将96只健康Wistar大鼠随机分为红景天治疗组、缺血再灌注组、假手术组和正常对照组。正常对照组未进行特殊处理,其他3组采用改良线栓法制成大脑中动脉缺血再灌注模型。观察各组神经功能缺损评分,2,3,5-三苯基氯化四唑染色测量各组脑梗死体积,采用免疫组织化学方法测定GFAP表达,用末端标记法测定神经细胞凋亡。结果红景天治疗组神经功能缺损评分低于缺血再灌注组(P<0.05),脑梗死体积小于缺血再灌注组(P<0.01)。GFAP阳性细胞于脑缺血2 h后即已出现,各组之间GFAP阳性表达比较差异无统计学意义。随缺血再灌注时间推移,缺血6~72 h时红景天治疗组较缺血再灌注组GFAP表达减低(P<0.05),红景天治疗组神经细胞凋亡较缺血再灌注组减少(P<0.05)。结论红景天胶囊能够减轻脑缺血再灌注损伤及提高神经功能评分,可能与减少星形胶质细胞GFAP的表达有关。
Objective To observe the effect of Hongjingtian capsule on the expression of glial fibrillary acidic protein (GFAP) in focal cerebral ischemia/reperfusion (I/R) injury rats. Methods 96 healthy Wistar rats were randomly divided into 4 groups: Hongjingtian group, I/R group, sham operation group and control group. The control group was processed without special treat- ment while the others were made into middle cerebral artery I/R models through the intra- luminal filament method. Neurological deficits were observed and infarct sizes were measured with 2, 3, 5-triphenyl-tetrazolium chloride staining. The expression alternations of GFAP were determined by immunohistochemical labeling method and the neuronal apoptosis was detected by terminal deoxynucleotidyl transferase mediated nick end labeling at corresponding hours. Re- sults Hongjingtian capsule group's neurological impairment have remarkable difference from I/R group's (P 〈0.05), and infarct sizes narrowed (P 〈0.01). The expression of GFAP appeared in 2 hours later, but not evidently. It increased gradually with the time of cerebral ischemia, and the expression of GFAP had become evident from 6 to 72 hours. The expression decreased in the Hongjingtian capsule groups than in I/R group (P 〈0.05) with the duration of I/R, the amount of apoptotie cells increased in the I/R group while only a small amount of apoptotic cells could be observed in the Hongjingtian group, and there were significant differences from I/R group (P 〈0.05). Cortclusiort Hongjingtian capsule can relieve ischemia reperfusion injury of brain tissue and improve neurologic scores, and by inhibiting the astroglial GFAP expression.
