粉防己碱对雄黄中毒大鼠尿砷血砷含量的影响 被引量:3

Effect of tetrandrine on arsenic content in rats with realgar poisoning

摘要 目的:观察粉防己碱(Tet)对雄黄中毒大鼠的排砷作用。方法:取大鼠60只,随机分为正常对照组、雄黄中毒模型组(模型组)、二巯基丙醇磺酸钠(DMPS)组、Tet高、中、低剂量组,每组10只。每组每日灌胃给予雄黄,各给药组分别给予不同剂量的药物腹腔注射,连续6周。给药期间,收集大鼠24 h尿以热消化-二乙氨基二硫代甲酸银(DDC-Ag)比色法的改良法测定尿砷含量,每周一次。停药后心脏取血用同样方法测定血砷含量。结果:各给药组大鼠24 h尿砷含量与模型组比较,Tet高、中剂量组以及低剂量组前4周的尿砷差异有极显著性(P均<0.01),低剂量组的第5、第6周尿砷量也有显著性差异(P<0.05);与DMPS组比较,仅低剂量组显示差异(P<0.05)。各给药组大鼠血砷含量与模型组比较,具有极显著性差异(P均 <0.01);与DMPS组比较,仅低剂量组有显著性差异(P<0.05)。结论:Tet对雄黄中毒大鼠具有明显的促进砷排泄作用。其作用与剂量呈正比例关系。 OBJECTIVE To investigate effects of tetrandrine (Tet) on eccritic function of arsenic in rats with realgar intoxication. METHODS Sixty rats were randomly divided into normal control group arsenic poisoning group ( model group ), arsenic poisoning treated by sodium dimercaptopropane sulfonate (DMPS) group (DMPS), arsenic poisoning treated by Tet high dosage group (Tet high dose group),middle dosage group (Tet middle dose group ) and low dosagee group (Tet low dose group) 10 rats (in each group. The realgar was taken by garage (ig) for each group once a day for 6 weeks except the normal control group. At the same time, drugs were taken by intraperitoneal injection (ip) for 6 weeks. During the drugs administrated, the each group rats' 24 h urine was collected to determine urine arsenic content by improved DDC-Ag way once a week. Blood arsenic content taken from the rats' heart was determined by the same method while the drugs were stopped administrating. RESULTS The 24 h urine arsenic content in the Tet high dose group, middle dose groups and low dose group show a very significant difference compared with the model group(all P〈0. 01), and hadn't statistical significance compared with the DMPS group except the Tet low dose group (P〈0. 05). The blood arsenic content in the 3 Tet doses groups also showed a very significant difference compared with the model group(all P〈0.01 ), and was a significant difference only in the Tet low dose group compared with the DMPS group(P〈0. 05). CONCLUSION It was demonstrated that Tet has outstanding promoting arsenic excretory effect in rats with realgar poisoning. Tet is an effective therapy with dose dependent potency for realgar intoxication.
